Expression and purification of polyhistidine-tagged firefly luciferase in insect cells

2001

The coleopteran firefly, Photinus pyralis, luciferase was produced in lepidopteran Trichoplusia ni insect cells using a baculovirus expression vector. The recombinant protein was equipped with a polyhistidine affinity tag at the carboxyl terminus and purified by immobilized metal-ion affinity chromatography in combination with an expanded bed adsorption system. This approach enabled an efficient, one-step purification protocol of a genetically modified luciferase with properties similar to those of the authentic counterpart. According to light emission measurements, the final yield of highly purified protein was 23 mg l−1 of cell culture. In addition, no specific interaction of interfering …

Detecting the stimulated decay of axions at radio frequencies

2018

Assuming axion-like particles account for the entirety of the dark matter in the Universe, we study the possibility of detecting their decay into photons at radio frequencies. We discuss different astrophysical targets, such as dwarf spheroidal galaxies, the Galactic Center and halo, and galaxy clusters. The presence of an ambient radiation field leads to a stimulated enhancement of the decay rate; depending on the environment and the mass of the axion, the effect of stimulated emission may amplify the photon flux by serval orders of magnitude. For axion-photon couplings allowed by astrophysical and laboratory constraints(and possibly favored by stellar cooling), we find the signal to be wi…

Doping induced stable room temperature dual emission from gadolinium doped vacancy ordered double halide perovskite, Gd:Cs2SnCl6

2022

The recent advancement in bandgap engineering through controlled doping has widen the prospectof vacancy ordered double halide perovskites (VO DHPs) by conferring them with designableoptoelectronic properties. Here, we report synthesis of Gd doped Cs2SnCl6 via a simplesolvothermal method. Gd3+ ions doping lowers the band gap from 3.8 eV to 2.8 eV and facilitatesstable room temperature dual PL emission centered at 440 nm and 610 nm. The macroscopicemission process is well supported by the confocal PL emission studies on isolated crystallites. Both the pristine (Cs2SnCl6) and Gd:Cs2SnCl6 exhibit crystalline cubic structure with Fm3m spacegroup. Rietveld refinement correlates well with the cubi…

Characterization of a neutron–beta counting system with beta-delayed neutron emitters

2016

A new detection system for the measurement of beta-delayed neutron emission probabilities has been characterized using fission products with well known β-delayed neutron emission properties. The setup consists of BELEN-20, a 4π neutron counter with twenty 3He proportional tubes arranged inside a large polyethylene neutron moderator, a thin Si detector for β counting and a selftriggering digital data acquisition system. The use of delayed-neutron precursors with different neutron emission windows allowed the study of the effect of energy dependency on neutron, β and β-neutron rates. The observed effect is well reproduced by Monte Carlo simulations. The impact of this dependency on the accura…

Search for intermediate mass black hole binaries in the first and second observing runs of the Advanced LIGO and Virgo network

2019

Gravitational wave astronomy has been firmly established with the detection of gravitational waves from the merger of ten stellar mass binary black holes and a neutron star binary. This paper reports on the all-sky search for gravitational waves from intermediate mass black hole binaries in the first and second observing runs of the Advanced LIGO and Virgo network. The search uses three independent algorithms: two based on matched filtering of the data with waveform templates of gravitational wave signals from compact binaries, and a third, model-independent algorithm that employs no signal model for the incoming signal. No intermediate mass black hole binary event was detected in this sear…

All-sky search in early O3 LIGO data for continuous gravitational-wave signals from unknown neutron stars in binary systems

2021

Rapidly spinning neutron stars are promising sources of continuous gravitational waves. Detecting such a signal would allow probing of the physical properties of matter under extreme conditions. A significant fraction of the known pulsar population belongs to binary systems. Searching for unknown neutron stars in binary systems requires specialized algorithms to address unknown orbital frequency modulations. We present a search for continuous gravitational waves emitted by neutron stars in binary systems in early data from the third observing run of the Advanced LIGO and Advanced Virgo detectors using the semicoherent, GPU-accelerated, binaryskyhough pipeline. The search analyzes the most s…

Anaerobic biodegradation of solvents from the packaging industry: study and enhancement

2017

Authentication of the protected designation of origin horchata de Valencia through the chemometric treatment of mineral content

2010

Spanish horchata de chufa samples were authenticated based on the determination of mineral elements by inductively coupled plasma atomic emission spectrometry (ICP-OES) combined with different chemometric methods. The ability of multivariate analysis, such as principle components analysis (PCA), classification and regression trees (CARTs) and discriminant analysis (DA) were evaluated in order to achieve a correct sample classification. It was possible to clearly differentiate homemade and long-life commercial samples by all three methods and CART and DA provided an excellent tool to establish the growth origin of the tiger nuts. CART analysis employed the concentration of Mg to discriminate…

Effect of organic matrices on the determination of the trace element chemistry (Mg, Sr, Mg/Ca, Sr/Ca) of aragonitic bivalve shells (<i>Arctica …

2010

The element chemistry of biogenic carbonates can provide important data on past environments. However, the Sr/Ca and Mg/Ca ratios as well as the Mg and Sr concentrations of biological carbonates, especially aragonitic bivalves often depart from apparent thermodynamic equilibrium. When measured in situ by means of LA-ICP-MS, the Mg concentration is often substantially enriched (two- to threefold) near the organic-rich, annual growth lines. To test the hypothesis that some organic components exert a major influence on the skeletal metal content, the element chemistry of different shell components (insoluble organic matrix, IOM; dissolved CaCO3 and soluble organics, SOM) of Arctica islandica w…

The role of the Krauss's organ in sound production in Pamphagidae (Caelifera: Orthoptera)

2012

Abstract In Pamphagidae, the Krauss's organ is located on both sides of the 2nd tergite and its function is superficially known. It has been proposed to have a stridulatory function. This organ can be found in all Pamphagidae except the apterous genera. Rearing specimens of some species and observing mounted pamphagids allowed the author to observe that when the Krauss's organ is absent (most apterous species) the tympanum is also absent; further, he discovered specialized stridulatory structures on hind femurs of some taxa for sound emission. By rearing two Pamphagidae belonging to different genera, fragments of stridulations involving the Krauss's organ and specialized structures on the h…
