Genome-wide characterization of local cattle breeds from central and western Mediterranean

Following-up on previous studies that investigated genetic relationships among several world-wide cattle breeds using genome-wide SNP data, we focus here on the central and western part of the Mediterranean. Detection of genomic signatures of selection in the North African Brune de l’Atlas cattle

The identification of signatures of selection in indigenous livestock genome is able to elucidate the adaptive mechanisms allowing them to cope with environmental pressures. In the present study, we used genotyping data from 106 Brune de l’Atlas cattle individuals and tests based on the extent of haplotype homozygosity (EHH) to look for footprints of natural selection. We identified 17 genomic regions displaying strong signal of selection (p-value <10-6) overlapping with previously described QTL regions affecting many traits such as meat and milk composition, tuberculosis susceptibility, tick and mastitis resistance. The “Caciocavallo Palermitano” is a typical stretched curd (pasta filata) cheese, made mainly in the Palermo area, in the Western Sicily, from cow milk. The aim of this investigation was to verify the influence of the on-farm production system (traditional vs. intensive) on some qualitative characteristics of “Caciocavallo Palermitano” cheese, with particular regard to fatty acid (FA) profile. The only population of the pseudodiaptomid copepod Calanipeda aquaedulcis Kritschagin, 1873 to date reported to occur in Sicily disappeared at the beginning of the XXI century due to deep environmental changes which affected the single site (Lake Biviere di Gela) known for this species on the island. In that site C. aquaedulcis is now replaced by Copidodiaptomus numidicus (Gurney, 1909), a diaptomid copepod whose distribution has been greatly increasing since the second half of the last century. Studies on methylation of PTHrP gene promoter 2 performed on five CpG island internal sites using the Methylation-Sensitive Restriction Endonuclease Multiplex (MSREM)-PCR showed that one of the sites nearest the trascription starting point is heavy methylated in a significantly high number of RA patients. Thus RA seems to be characterized by genomewide hypomethylation associated with local hypermethylation like the most part of tumors. This result raises the possibility that susceptibility to lymphomas is related to abnormal DNA methylation levels and suggests the opportunity to evaluate the DNA methylation status in RA patientin fact the demethylating therapies together with diet and life style can act towards an increase of tumor risk. Future studies using a larger number of subjects could confirm these findings.Rheumatoid Arthritis (RA) is a chronic multisystem inflammatory disease characterized by high recurrence of lymphomas as well as hypercalcemia due to PTHrP overexpression. Because of DNA methylation plays a critical role in development of neoplasias we determined in RA patients the global DNA methylation status and local methylation pattern of the CpG island of one of the three promoters of PTHrP gene utilizing molecular and cytogenetic techniques. Investigations performed on DNA from peripheral blood of patients and donors amplified by Methylation-Sensitive Arbitrarily Primed (MeS-AP)-PCR indicated that RA is strongly associated with global DNA hypomethylation. Similarly chromosomal DNA methylation pattern analysis by indirect immunofluorescence technique with anti 5-methylcitosine antibody showed all peripheral lymphocyte metaphases from RA patients with chromosomes weakly fluorescent without discrete grain

This contribution illustrates the preliminary results of our Research Unit in the PRIN Project 2010-11, which focuses on active and recent geodynamics of Calabrian Arc. The integration of the new geological, geodetic and seismological data supports the inferred recent plate boundary reorganization in the central-southern Mediterranean, where the regional GNNS velocity fields point to a deceleration or cessation of Calabrian Arc migration, and to extension along the axis of the Calabrian Arc, accommodated by normal faulting (e.g. Capo Vaticano and Messina Straits (Aloisi et al., 2012; Pepe et al., 2014; Spampinato et al., 2014).
