Cyclic GMP (cGMP) formation in rat pinealocytes is regulated through a synergistic dual receptor mechanism involving beta- and alpha 1-adrenergic receptors. The effects of NG-monomethyl-L-arginine (NMMA), which inhibits nitric oxide (NO) synthase and NO-mediated activation of cytosolic guanylate cyclase, and methylene blue (MB), which inhibits cytosolic guanylate cyclase, were investigated in an attempt to understand the role of NO in adrenergic cGMP formation. Both NMMA and MB inhibited beta-adrenergic stimulation of cGMP formation as well as alpha 1-adrenergic potentiation of beta-adrenergic stimulation of cGMP formation, whereas they had no effect in unstimulated pinealocytes. Anderson-Fabry disease, an inherited deficiency in the lysosomal enzyme alpha-galactosidase A, is characterized by the progressive accumulation of globotriaosylceramide (Gb3), also known as CD77. We sought to clarify the pathogenesis of Fabry disease by establishing a cell model of this disorder. The expression of alpha-galactosidase A was transiently silenced by RNA interference in HK2 and primary human renal epithelial cells and stably silenced in HK2 cells by retroviral transfection with small hairpin RNA. Abstract We classify finite-dimensional real nilpotent Lie algebras with 2-dimensional central commutator ideals admitting a Lie group of automorphisms isomorphic to SO 2 ( ℝ ) ${{\mathrm{SO}}_{2}(\mathbb{R})}$ . This is the first step to extend the class of nilpotent Lie algebras 𝔥 ${{\mathfrak{h}}}$ of type { n , 2 } ${\{n,2\}}$ to solvable Lie algebras in which 𝔥 ${{\mathfrak{h}}}$ has codimension one.

Modal Consequence Relations Extending S4.3: An Application of Projective Unification

We characterize all finitary consequence relations over $\mathbf{S4.3}$ , both syntactically, by exhibiting so-called (admissible) passive rules that extend the given logic, and semantically, by providing suitable strongly adequate classes of algebras. This is achieved by applying an earlier result stating that a modal logic $L$ extending $\mathbf{S4}$ has projective unification if and only if $L$ contains $\mathbf{S4.3}$ . In particular, we show that these consequence relations enjoy the strong finite model property, and are finitely based. RAB18 Loss Interferes With Lipid Droplet Catabolism and Provokes Autophagy Network Adaptations

Autophagy is dependent on appropriate lipid supply for autophagosome formation. The regulation of lipid acquisition and the autophagy network response to lipid-limiting conditions are mostly elusive. Here, we show that the knockout of the RAB GTPase RAB18 interferes with lipid droplet catabolism, causing an impaired fatty acid release. The resulting reduced lipid-droplet-derived lipid availability influences autophagy and provokes adaptive modifications of the autophagy network. These adjustments include increased expression and phosphorylation of ATG2B as well as augmented formation of the ATG12-ATG5 conjugate.
