Problems with primary vs. secondary grammaticalization: the case of East and mainland Southeast Asian languages

2015

Abstract The present paper aims at testing four criteria for the distinction between primary vs. secondary grammaticalization in East and mainland Southeast Asian languages. These languages are of special interest for research on grammaticalization because they show limitations in the coevolution of meaning and form and because pragmatic inference is very prominent (lack of obligatory grammatical markers, multifunctionality). If the four criteria work in these languages, this can be taken as a good indicator for their cross-linguistic, maybe universal relevance. The criteria are tested with three markers that stand for three different types of multifunctionality: (i) the Khmer verb baːn ‘co…

A diachronic study of the (negative) additive «anche» in italian

2016

Evidential markers derived from visual perception in Spanish dialects : a contrastive study of por lo visto and se ve (que)

2019

Verbs of perception (PV) are known to frequently give rise to discursive uses. PVs are used, for example, as evidential markers through grammaticalization processes. This study focuses on two evidential markers of Spanish derived from the visual PVs ver ('to see'), namely, 'por lo visto' and 'se ve (que)'. Although the attention dedicated to these evidential markers in Spanish has increased considerably in recent years, contrastive studies have not taken into account the dialectal variation of this phenomenon. We aspire to take a first step in filling this gap by contrastively studying 'por lo visto' and 'se ve (que)' in different dialects of Spanish by means of a comprehensive, empirical s…

Secondary grammaticalization and the English adverbial -ly suffix

2015

Author's version of an article in the journal: Language sciences. Also available from the publisher at: http://dx.doi.org/10.1016/j.langsci.2014.10.003 This paper discusses the secondary grammaticalization of the English adverbial -ly suffix and makes claims about the concept of secondary grammaticalization. Secondary grammaticalization is defined as the development of a new grammatical function in an already grammatical element. It is shown that the development of the -ly suffix involves a number of the processes which are associated with grammaticalization, e.g. paradigmatization, specialization, obligatorification, subjectification, layering and persistence. However, none of these proces…

Mitochondrial G protein coupled receptor kinase 2 regulates proinflammatory responses in macrophages.

2013

G-protein-coupled receptor kinase 2 (GRK2) levels are elevated in inflammation but its role is not clear yet. Here we show that GRK2 expression is dependent on NFκB transcriptional activity. In macrophages, LPS induces GRK2 accumulation in mitochondria increasing biogenesis. The overexpression of the carboxy-terminal domain of GRK2 (βARK-ct), known to displace GRK2 from plasma membranes, in macrophages induces earlier localization of GRK2 in mitochondria in response to LPS leading to increased mt-DNA transcription, reduced ROS production and cytokines expression. Our study shows the relevance of GRK2 subcellular localization in macrophage’s biology and its potential therapeutic properties i…

Colocalization but differential regulation of neuronal NO synthase and nicotinic acetylcholine receptor in C2C12 myotubes.

2003

In mammalian skeletal muscle, neuronal-type nitric oxide synthase (nNOS) is found to be enriched at neuromuscular endplates. Here we demonstrate the colocalization of the nicotinic acetylcholine receptor (nAChR, stained with α-bungarotoxin) and nNOS (stained with a specific antibody) in murine C2C12myotubes. However, coimmunoprecipitation experiments demonstrated no evidence for a direct protein-protein association between the nAChR and nNOS in C2C12myotubes. An antibody to the α1-subunit of the nAChR did not coprecipitate nNOS, and an nNOS-specific antibody did not precipitate the α1-subunit of the nAChR. Treatment of mice with bacterial LPS downregulated the expression of nNOS in skeletal…

Nanoscale distribution of TLR4 on primary human macrophages stimulated with LPS and ATI

2019

Toll-like receptor 4 (TLR4) plays a crucial role in the recognition of invading pathogens. Upon activation by lipopolysaccharides (LPS), TLR4 is recruited into specific membrane domains and dimerizes. In addition to LPS, TLR4 can be stimulated by wheat amylase-trypsin inhibitors (ATI). ATI are proteins associated with gluten containing grains, whose ingestion promotes intestinal and extraintestinal inflammation. However, the effect of ATI vs. LPS on the membrane distribution of TLR4 at the nanoscale has not been analyzed. In this study, we investigated the effect of LPS and ATI stimulation on the membrane distribution of TLR4 in primary human macrophages using single molecule localization m…

An international genome-wide meta-analysis of primary biliary cholangitis: Novel risk loci and candidate drugs.

2021

[BACKGROUND & AIMS] Primary biliary cholangitis (PBC) is a chronic liver disease in which autoimmune destruction of the small intra-hepatic bile ducts eventually leads to cirrhosis. Many patients have inadequate response to licensed medications, motivating the search for novel therapies. Previous genome-wide association studies (GWAS) and meta-analyses (GWMA) of PBC have identified numerous risk loci for this condition, providing insight into its aetiology. We undertook the largest GWMA of PBC to date, aiming to identify additional risk loci and prioritise candidate genes for in silico drug efficacy screening. [METHODS] We combined new and existing genotype data for 10, 516 cases and 20, 77…

Apprentissage de modalités auxiliaires pour la localisation basée vision

2018

In this paper we present a new training with side modality framework to enhance image-based localization. In order to learn side modality information, we train a fully convo-lutional decoder network that transfers meaningful information from one modality to another. We validate our approach on a challenging urban dataset. Experiments show that our system is able to enhance a purely image-based system by properly learning appearance of a side modality. Compared to state-of-the-art methods, the proposed network is lighter and faster to train, while producing comparable results.
