Ability of the T cell-replacing polyanion dextran sulfate to trigger the alternate pathway of complement activation.

Dextran sulfate (DS) consumed C3 in C4 deficient guinea pig serum. This temperature-dependent reaction required Mg++ ions and could therefore be blocked by EDTA. Isolated C3 was not influenced by DS, but serum factors were required for C3 consumption. The C3 proactivator as well as C3 were converted to their activated state by DS in guinea pig and human serum, as revealed by immunoelectrophoretical analysis. DS generated anaphylatoxin activity in serum. It is concluded that DS activates C3 via the alternate pathway of complement activation. Current concepts of activation of the alternative pathway of complement (APC) focus on the central role of an amplification mechanism triggered by C3b which is covalently bound to the surfact of activating substances. Using sulfated polyanions as model substances, an efficient fluid-phase activation of complement is demonstrated in contrast to solid-phase activation. It is shown that particulate high-molecular weight sulfated polyanions are capable of reversible binding the guinea pig and human regulatory protein beta1H. Migration of Leukocytes into Filters Coated Homogeneously with Immune Complexes, Antigens, Lectins or Tripeptides

Cellulose nitrate filters were incubated in solutions of albumin, a chemotactically active tripeptide (f-Met-Leu-Phe), immune complexes or lectins and afterwards washed with buffer. They showed a dose-dependent increased leukocyte migration, when tested in typical Boyden chambers in comparison to filters treated only with buffer. The tripeptide, the immune complexes and the lectins were stimulatory at very low concentrations and acted inhibitory at high concentrations. Presence of potentially pathogenic free-living amoebae strains from well water samples in Guinea-Bissau

Free-living amoebae (FLA) include opportunistic pathogens such as Naegleria fowleri, Balamuthia mandrillaris, and the genera Sappinia and Acanthamoeba. In this study, a survey was conducted in order to evaluate the presence of potentially pathogenic amoebic strains in water samples collected from wells located in the western part of Guinea-Bissau. The samples were left to precipitate for 48 hours and then the sediments were seeded on non-nutrient agar plates containing Escherichia coli spread and cultures were checked daily for the presence of FLA. Potassium antimonate was used to precipitate calcium in the cochlea of the guinea-pig. The distribution of the calcium antimonate precipitates was analysed by electron microscopy. The precipitate density was determined in different cell types in the organ of Corti by counting the number of calcium binding sites in a 10-micron 2 area. The size of the precipitates varied considerably, and thus the relative amount of the precipitable calcium was estimated only semi-quantitatively. Use of electron spectroscopic imaging to determine element composition of the melanin granules in the stria vascularis of the guinea pig.

Electron spectroscopic imaging (ESI) was used to analyze the element content of melanin granules in the stria vascularis seen in ultrathin sections of Spurr-embedded cochleae of the guinea pig. To determine element composition, ESI images were taken at different ionization edges, and non-specific background signals were subtracted digitally by an image processing system. The presence of calcium and nitrogen in the melanin granules could be demonstrated clearly. Antispasmodic effect of 4'-methylepigallocatechin on guinea pig ileum.

AbstractThe antispasmodic effect of 4′-methylepigallocatechin (MEC), which was isolated from Maytenus rigida Mart (Celestraceae), was investigated in vitro in guinea pig intestinal segments. In the isolated ileum, MEC (1 nM–100μM) did not modify the ileal spontaneous tonus or the electrically elicited contractions. MEC (8μM) significantly (p<0.01) reduced the submaximal contractions induced by histamine (2μM), carbachol (100μM) and BaCl2 (0.03M). An additive relaxing action (p<0.001) was observed by co-incubation of verapamil (10 nM) and MEC (8μM).
