Host-directed therapies for COVID-19

Purpose of review Severe acute respiratory syndrome coronavirus-2-induced hyperinflammation is a major cause of death or end-organ dysfunction in COVID-19 patients. We review adjunct host-directed therapies (HDTs) for COVID-19 management. Recent findings The use of umbilical cord-derived mesenchymal stem cells as HDT for COVID-19 has been shown to be safe in phase 1 and 2 trials. Trials of anti-interleukin-6 receptor antibodies show promising mortality benefit in hospitalized COVID-19 patients. Repurposed drugs and monoclonal antibodies targeting specific cytokines acting on different aspects of the pro- and anti-inflammatory cascades are under evaluation. Despite the existence of treatment for diabetes, inadequate metabolic control triggers the appearance of chronic complications such as diabetic retinopathy. Diabetic retinopathy is considered a multifactorial disease of complex etiology in which oxidative stress and low chronic inflammation play essential roles. Chronic exposure to hyperglycemia triggers a loss of redox balance that is critical for the appearance of neuronal and vascular damage during the development and progression of the disease. Cardiovascular diseases (CVD) are the clinical manifestation of atherosclerosis, a chronic inflammatory disease promoted by several risk factors such as dyslipidemia, type 2 diabetes mellitus, hypertension, and smoking. Acute CVD events are the result of an unresolved inflammatory chronic state that promotes the rupture of unstable plaque lesions. Conduit dynamics and post explosion degassing on Stromboli: A combined UV camera and numerical modeling treatment.

Abstract Recent gas flux measurements have shown that Strombolian explosions are often followed by periods of elevated flux, or “gas codas,” with durations of order a minute. Here we present UV camera data from 200 events recorded at Stromboli volcano to constrain the nature of these codas for the first time, providing estimates for combined explosion plus coda SO2 masses of ≈18–225 kg. Numerical simulations of gas slug ascent show that substantial proportions of the initial gas mass can be distributed into a train of “daughter bubbles” released from the base of the slug, which we suggest, generate the codas, on bursting at the surface. Abstract How do configurations of humans and algorithms evolve as firms adopt artificial intelligence (AI) capabilities, and what are the implications for work and organization? We explored these questions through a two-year long case study of an organization in the international maritime trade that introduced automated algorithmic support for data analysis and prediction work. Drawing on a human–machine configuration perspective, we found that humans and the algorithm were configured and reconfigured in multiple ways over time as the organization dealt with the introduction of algorithmic analysis. Lead optimization through VLAK protocol: New annelated pyrrolo-pyrimidine derivatives as antitumor agents

Abstract The chemometric protocol VLAK was applied to predict improvement of the biological activity of pyrrolo-pyrimidine derivatives as anticancer agents, by using the NCI ACAM Database as depository of antitumor drugs with a known mechanism of action. Among the selected compounds two of these showed a good increase in the antitumor activity. These new pyrrolo-pyrimidine compounds were demonstrated effective against the full panels of NCI DTP tumour human cell lines. Abstract Uncertainty and the pursuit of new ventures are intricately linked. Digital technologies open up new ways for uncertainty handling in the pursuit of novelty. In this paper, we develop a process model that traces the dynamics among uncertainty, digitally enabled tactics, and organizational novelty. The model points to the potential of technologies that are evocative, disposable, and responsive for venturing in complex, uncertainty-ridden contexts.
