Antidiabetic Behavior of Biguanides

The existence of active electron pairs on some nitrogen atoms in phenformin hydrochloride is inferred from the presence of a hydrogen catalytic polarographic wave. This finding emphasizes the ability of biguanides to form hydrogen bridges with other molecular species such as amino acids and proteins, as well as to form coordination complexes with zinc and other metallic cations by means of these electron pairs. The antidiabetic action of phenformin and other related biguanides can be explained in terms of competition between these molecules and insulin to coordinate cationic oligoelements together with their ability to form hydrogen bonds between the biguanide moiety and insulin itself.

Reactive oxygen species induce oxidative modification of critical macromolecules. Oxygen derived free radicals may act as potential cytotoxic intermediates inducing inflammatory and degenerative processes, or as signal messengers for the regulation of gene expression. Livelli serici di insulina nel soggetto normale (Obeso e non) e nel diabetico obeso Dopo Carico Intragastrico di Aminoacidi

Gli AA. hanno somministrato in 5 soggetti normali normosomici, in 5 obesi metabolicamente sani e in 5 diabetici obesi un carico intragastrico (mediante sondino) di 9 aminoacidi essenziali (arginina, fenil-alanina, valina, leucina, lisina, metionina, isoleucina, istidina, triptofano) per un totale di g 30 ed hanno valutato la risposta insulo-pancreatica. In tutti i casi di soggetti in esame si e messo in evidenza un significativo aumento dell'insulinemia, determinata con metodo radioimmunologico. Background: Little is known about the association between adrenarche and cognition in general populations of children. We therefore studied the associations of dehydroepiandrosterone sulfate (DHEAS), androstenedione (A4), testosterone, insulin-like growth factor-1 (IGF-1), and adrenarche with cognition among prepubertal children.
