In Vivo and In Vitro Binding of Vip3Aa to Spodoptera frugiperda Midgut and Characterization of Binding Sites by 125 I Radiolabeling

ABSTRACT Bacillus thuringiensis vegetative insecticidal proteins (Vip3A) have been recently introduced in important crops as a strategy to delay the emerging resistance to the existing Cry toxins. The mode of action of Vip3A proteins has been studied in Spodoptera frugiperda with the aim of characterizing their binding to the insect midgut. Immunofluorescence histological localization of Vip3Aa in the midgut of intoxicated larvae showed that Vip3Aa bound to the brush border membrane along the entire apical surface. ABSTRACT It is generally accepted that Bacillus thuringiensis Cry toxins insert into the apical membrane of the larval midgut after binding to specific receptors, and there is evidence that the distribution of binding molecules along the midgut is not uniform. By use of the voltage-sensitive dye DiSC 3 (5) and 125 I-labeled Cry1Ac, we have measured the effect of Cry1Ac in terms of permeabilization capacity and of binding parameters on brush border membrane vesicles (BBMV) prepared from the anterior and the posterior regions of the larval midgut from two insect species, Manduca sexta and Helicoverpa armigera . Stereoselective syntheses of piperidinones and their modification by organometallic coupling reactions

Dehydropiperidinones stereoselectively obtained from N-arabinosyl imines were iodinated at the enaminone structure. Knochel iodine-magnesium exchange afforded Grignard compounds of these piperidinone derivatives which reacted, either directly or after transmetalation to zinc or copper intermediates, with alkyl-, aryl- or acylhalides to give correspondingly substituted piperidinones. Stereoselective conjugate allyl cuprate addition to a thus obtained 5-allyl dehydropiperidinone and ring-closing metathesis of the product gave a hydroquinolinone containing three stereogenic centers.

Quality control of commercial mustard by thin-layer chromatography

Horseradish samples ( Cochlearis armoracia L. ) and mustard samples ( Sinapis alba L. ) were extracted with 70:30 ( v/v ) methanol-water in accordance with ISO method 91671. Samples of commercial mustard with and without horseradish were submitted to the same extraction procedure. Speciation of tetraalkyllead compounds by flow injection — atomic absorption spectrophotometry

The on-line demetallation of alkyllead compounds, with iodine, and further emulsification, provides a fast determination of total lead content in liposoluble matrices, such as gasolines. It can be carried out in a double channel manifold, using a magnetically well stirred dilution chamber for the emulsification of the samples, previous to their analysis by flame atomic absorption spectrophotometry. On the other hand, the different behaviour of tetraethyllead (TEL) and tetramethyllead (TML) can be employed for speciation of both compounds in the sample. Corticotropin-releasing factor (CRF) is thought to be involved in the regulation of the diurnal activity of the hypothalamus-pituitary-adrenal (HPA) axis and to act as a neurotransmitter in the brain. Observations of high concentrations of I2and IO in coastal air supporting iodine-oxide driven coastal new particle formation

[1] Theoretical studies have predicted that concentrations of gaseous I2 and IO of the order of 80–100 ppt and 40–50 ppt, respectively, are required in coastal air to account for photochemically-driven coastal new-particle formation events to occur. However, measurements reported to date (i.e., ∼20 ppt I2, ≤ 10 ppt IO) have not supported the required model predictions. Here, we present measurements of high concentrations of I2 and IO in N.E. Atlantic marine air on the west coast of Ireland. The maximum mixing ratios of daytime I2 and IO over the seaweed beds during low tide were 302 ppt and 35 ppt, respectively. INFLUENCE OF INTRA-CORONARY ENHANCEMENT ON DIAGNOSTIC ACCURACY WITH 64-SLICE CT CORONARY ANGIOGRAPHY

We assessed the effect of intra-coronary attenuation on diagnostic accuracy using 64-slice computed tomography coronary angiography (CT-CA). We enrolled 170 patients with suspected coronary artery disease who underwent conventional coronary angiography (CA) and 64-slice CT-CA (100 ml of Iomeprol 400 mg I/ml at 4 ml/s). The study population was divided into two groups (85 patients each based on median attenuation of 326 HU) based on mean arterial attenuation; group 1 with low attenuation and group 2 with high attenuation.
