In this study, the mucilage polysaccharide (MP) from Amanita hemibapha subspecies javanica was prepared by hot water extraction and ethanol precipitation and then fractionated using anion-exchange chromatography equipped with a DEAE Sepharose fast flow column. The most immune-enhancing polysaccharide fraction 2 (MPF2) was subjected to a structural modification such as hydrolysis or over-sulphation. The sulphate and molecular weight (Mw) of over-sulphated (OS1-3) and hydrolysed (HS1-3) derivatives of MPF2 differed between 9.85% and 14.2% and 32.8 and 88.1 × 103 g/mol, respectively. Effect of Lignin-Containing Media on Growth of Medicinal Mushroom Lentinula Edodes

Abstract The effect of lignocellulose and lignin on growth of mycelium of mushroom Lentinula edodes and laccase activity in cultivation medium was studied. It was shown that cultivation of L. edodes mycelium in liquid nutrient medium with addition of 0.25-0.5% of kraft lignin increased mycelium biomass yield approximately two times compared with reference conditions without addition of lignin. Similar results were obtained in experiments in which 0.5% lignocellulose that remained after obtaining furfural, and 0.5% lignin that remained after obtaining furfural and glucose from wheat straw, were added to the nutrient medium. Biogeneration of 1-octen-3-ol by lipoxygenase and hydroperoxide lyase activities of Agaricus bisporus

Abstract The biogeneration of 1-octen-3-ol by lipoxygenase and associated hydroperoxide lyase activities of a homogenate of the cultivated mushroom Agaricus bisporus was investigated using linoleic acid as a substrate. The optimal substrate and protein concentrations were 1.5 mM and 1.5 mg ml −1 , respectively. The V max value for the enzymic coupled reaction lipoxygenase–hydroperoxide lyase was 6 μg of 1-octen-3-ol ml −1 min −1 , with a K m value of 0.3×10 −3 M. The highest enzymic activity was observed in the presence of pure oxygen (22.9 μg of 1-octen-3-ol ml −1 ), with a bioconversion yield of 36%. Abstract The fruiting bodies of fungi sprout from mycelium are capable of accumulating significant amounts of trace elements, both metals and metalloids. Content of these elements in fruiting bodies may exceed their concentration in the substrate where fungi develop. Among the elements the radioactive nuclides are also present. In this work health risk caused by increased radioactivity dose absorbed with Xerocomus badius bay bolete consumption was estimated. Simultaneous Determination of Thiamin and Riboflavin in Mushrooms by Liquid Chromatography

A simple, fast, inexpensive, and reliable method useful for the simultaneous, routine determination of thiamin and riboflavin in mushrooms is examined. It uses the extraction procedure, with slight modifications, proposed by the AOAC for the extraction of thiamin and riboflavin, followed by a liquid chromatographic separation on a reversed-phase Spherisorb ODS column with methanol/water as mobile phase gradient. Four new Candida cretensis strains isolated from Spanish fermented sausages (chorizo): Taxonomic and phylogenetic implications

Four yeast strains were isolated from Spanish traditional fermented sausages (chorizo) spoiled by gas production. Using the classical identification procedures, they were identified as Debaryomyces hansenii. However, they fermented galactose and did not produce positive results in Debaryomyces differential medium (DDM), a growth medium highly specific for this species. Phylogenetic analysis showed identical sequences for the D1/D2 domain of the 26S rRNA gene and almost identical sequences for the 5.8S-ITS region with those of the recently described yeast species Candida cretensis. Abstract N6‐methyladenosine (m6A) regulates a variety of physiological processes through modulation of RNA metabolism. This modification is particularly enriched in the nervous system of several species, and its dysregulation has been associated with neurodevelopmental defects and neural dysfunctions. In Drosophila, loss of m6A alters fly behavior, albeit the underlying molecular mechanism and the role of m6A during nervous system development have remained elusive.
