Staphylococcal α-toxin is a 293-residue, single-chain polypeptide that spontaneously assembles into a heptameric pore in target cell membranes. To identify the pore-forming domain, substitution mutants have been produced in which single cysteine residues were introduced throughout the toxin molecule. By attaching the environmentally sensitive dye acrylodan to the sulfhydryl groups, the environment of individual amino acid side chains could be probed. In liposomes, a single 23-amino acid sequence (residues 118–140) was found to move from a polar to a nonpolar environment, indicating that this sequence forms the walls of the pore. This work investigated the oxidative injury to human red blood cells (RBCs) by the exposure to exogenous malondialdehyde (MDA), in a physiological environment. When a 10% RBC suspension was incubated in autologous plasma, in the presence of 50 microM MDA, 30% of MDA entered into the cells. A time-course study showed that MDA caused early (30-120 min) and delayed (3-18 h) effects. MDA caused a fast depletion of reduced glutathione, and loss of the glucose-6-phosphate dehydrogenase activity, followed by a decrease of HbO2. Accumulation of methemoglobin, and formation of small amounts of hemichrome were later evident. Also, an HbO2-derived fluorescent product was measured in the membrane. The …

The conductance of potassium iodide has been measured in the solvents ethylene carbonate, water, methyl ethyl ketone, and pairwise mixtures of these solvents at 40°C; and ethylene carbonate-water, tetramethylene sulfone-water, dimethyl sulfoxide-water, tetrahydrofuran-water, ethylene carbonate-tetramethylene sulfone-water, ethylene carbonate-tetramethylene sulfone, and tetrahydrofuran-dimethyl sulfoxide at 25°C. For dielectric constants greater than about 60, the pairing constants KA are in the range 0.3–2.0; no correlation between KA and solvent properties could be established. Abstract Background KCNE1 represents the regulatory beta-subunit of the slowly activating delayed rectifier potassium channel (IKs). Variants of KCNE1 have repeatedly been linked to the long-QT syndrome (LQTS), a disorder which predisposes to deafness, ventricular tachyarrhythmia, syncope, and sudden cardiac death. Abstract The chemiluminescent behaviour of the amino acridines like acriflavine, and proflavin is reported. Different strong oxidants (potassium permanganate, cerium(IV), hexacyanoferrate(III), hydrogen peroxide in different media) were tested, and potassium permanganate in sulphuric acid medium was selected. The study of the experimental parameters affecting the oxidation and detection was performed with the aid of a FIA assembly. Direct flow injection chemiluminescence determination of salicylamide

Abstract A new direct flow injection chemiluminescence method is proposed for the determination of salicylamide, based upon the oxidation of the drug by potassium permanganate in dilute sulphuric acid. The calibration graph is linear over the range 20 ng ml−1 (30 limit of detection)–8 μg ml−1 salicylamide, with a relative standard deviation (n=50, 0.5 μg ml−1) of 1.7%. The average sample insertion rate is 142 h−1. The influence of relevant foreign compounds is found to be relatively slight. The method is applied to the determination of salicylamide in a pharmaceutical formulation and human urine.
Direct determination of calcium, magnesium, sodium and potassium in water by flow injection flame atomic spectroscopy, using a dilution chamber

A simple procedure to carry out the direct analysis of calcium, magnesium, sodium and potassium in water by flow injection analysis (FIA) using flame atomic absorption spectroscopy (AAS) or flame photometry (FP) has been developed, using a well stirred dilution chamber to extend the calibration range, and both a double injector and the merging zone technique to add a lanthanum solution to samples and standards. The results obtained in the analysis of real samples agree with those found by a batch flame atomic procedure.
