A New Culture Technique: An Approach to the in-Vitro Reconstitution of Endocrine Organs

A new cell culture technique called three-dimensional-rotation culture (3dR-culture) has been developed. Its main characteristics are: (i) cultivation under constant rotation, and (ii) the use of a three-dimensional material that allows the cells to adhere and to grow in a three-dimensional arrangement. Two glassfibre materials and one gelatine-sponge proved suitable for the use in cell culture. The new technique has the advantage to yield in large numbers cells bound on a stable matrix that can easily be handled, e.g. for implantation.

Isolated neurointermediate lobes (NILs) or isolated neural lobes (NLs) of the rat pituitary gland were incubated in Krebs-HEPES solution which contained pargyline and the dopamine uptake inhibitor GBR 12921. The release of endogenous dopamine was determined by HPLC with electrochemical detection. Electrical stimulation of the pituitary stalk induced a frequency-dependent release of dopamine. The release of dopamine from the combined NIL evoked by stimulation at 15 Hz was increased by 130% in the presence of the dopamine D2 receptor antagonist, (-)-sulpiride; the (+)-enantiomer of sulpiride had virtually no effect. Isolated rat NILs were incubated in Krebs-HEPES solution. The release of dopamine and its metabolites (DOPAC, HVA and MOPET) was determined by HPLC with electrochemical detection. The spontaneous release of the sum of metabolites was about 40 times that of dopamine. The spontaneous outflow of dopamine metabolites was unaffected after inhibition of dopamine uptake (by GBR 12921) or after pretreatment with reserpine (5 mg/kg, 12 h before the experiments), but it was reduced by 50% after preincubation with the irreversible DOPA decarboxylase inhibitor, (MFMD, 10 microM, for 10 min). The effect of natural angiotensin converting enzyme inhibitors on kallikrein activity

SARS-Cov-2 vīruss, izmantojot angiotenzīna-konvertējošo enzīmu 2 (AKE2) kā receptoru, iekļūst šūnās un izmaina angiotenzīna un bradikinīna sistēmu līdzsvaru. Pacientiem slimības sākumā var parādīties plaušu tūska, kas ir saistīta ar vietēju asinsvadu caurlaidību. Inhibējot kallikreīna aktivitāti varētu samazināt bradikinīna sintēzi, kas samazinātu vīrusa izraisītās plaušu tūskas. Šī pētījuma mērķis ir pētīt dažu dabas vielu ar AKE2 inhibitoru īpašībām spēju inhibēt kallikreīna aktivitāti in vitro. Oligodendrocytes in mouse corpus callosum are coupled via gap junction channels formed by connexin47 and connexin32.

According to previously published ultrastructural studies, oligodendrocytes in white matter exhibit gap junctions with astrocytes, but not among each other, while in vitro oligodendrocytes form functional gap junctions. We have studied functional coupling among oligodendrocytes in acute slices of postnatal mouse corpus callosum. By whole-cell patch clamp we dialyzed oligodendrocytes with biocytin, a gap junction-permeable tracer. On average 61 cells were positive for biocytin detected by labeling with streptavidin-Cy3.
