Location and characterization of the stem-calyx area on oranges by computer vision

Three image analysis methods were studied and evaluated to solve the problem of removing long stems attached to mechanically harvested oranges: colour segmentation based on linear discriminant analysis, contour curvature analysis, and a thinning process which involves iterating until the stem becomes a skeleton. These techniques are able to determine the presence or absence of a stem with certainty, to locate the stems from random views with more than 90% accuracy and from profile images with an accuracy ranging from 92.4% to 100% depending on the method used. Seasonal-variation of nitrogenous compounds in the xylem sap of citrus

Samples of tracheal sap of Citrus sinensis (L.) Osbeck cv. Washington Navel were taken from field trees throughout the year and the nitrogen composition of the sap was determined. The nitrogenous fraction of the sap was composed mainly of free amino acids (92–97% of total nitrogen) and nitrates throughout the year. Proline was the most abundant amino acid during almost the entire cycle, and its concentration was especially high during the autumn and winter period. Nevertheless, a significant part (40–60%) of the total organic nitrogen was transported as arginine. Total nitrogen as well as amino acids and nitrates were maximal at spring flush. We describe the development of a high-performance liquid chromatography (HPLC) method for the determination of citrulline and other amino acids relevant to intestinal diseases. The amino acids were derivatized with 9-fluorenylmethylchloroformate (FMOC-Cl) and their derivatives were separated on two different columns, a core-shell column (Halo C18) and a silica-based monolith (Chromolith Performance RP-18). The derivatization reaction was optimized with respect to pH, buffer concentration and reproducibility. Fight against Citrus tristeza virus (CTV) implies the use of CTV-resistant rootstocks, such as citranges [C. Sinensis (L.) Osb. × P. Trifoliata (L.) Raf.]. The 'C35' represents a promising tolerant to CTV genotype.
