Increased AICD generation does not result in increased nuclear translocation or activation of target gene transcription.

2008

A sequence of amyloid precursor protein (APP) cleavages culminates in the sequential release of the APP intracellular domain (AICD) and the amyloid beta peptide (Abeta) and/or p3 fragment. One of the environmental factors favouring the accumulation of AICD appears to be a rise in intracellular pH. Here we further identified the metabolism and subcellular localization of artificially expressed constructs under such conditions. We also co-examined the mechanistic lead up to the AICD accumulation and explored possible significances for its increased expression. We found that most of the AICD generated under pH neutralized conditions is likely cleaved from C83. While the AICD surplus was unable…

Insensitivity to Aβ42-lowering Nonsteroidal Anti-inflammatory Drugs and γ-Secretase Inhibitors Is Common among Aggressive Presenilin-1 Mutations

2007

Abeta42-lowering nonsteroidal anti-inflammatory drugs (NSAIDs) constitute the founding members of a new class of gamma-secretase modulators that avoid side effects of pan-gamma-secretase inhibitors on NOTCH processing and function, holding promise as potential disease-modifying agents for Alzheimer disease (AD). These modulators are active in cell-free gamma-secretase assays indicating that they directly target the gamma-secretase complex. Additional support for this hypothesis was provided by the observation that certain mutations in presenilin-1 (PS1) associated with early-onset familial AD (FAD) change the cellular drug response to Abeta42-lowering NSAIDs. Of particular interest is the P…

Aberrant splicing of the Drosophila melanogaster phenylalanine hydroxylase pre-mRNA caused by the insertion of a B104/roo transposable element in the…

1999

Abstract We report the insertion of the transposable element B104 in the Phenylalanine hydroxylase gene of the Drosophila mutant Henna-recessive 3 . Its presence alters the Phenylalanine hydroxylase splicing pattern, producing at least two aberrant mRNAs which contain part of the B104 sequence interrupting the coding region. This aberrant splicing is provoked by the use of a cryptic donor site encoded by the B104 3′ long terminal repeat in combination with either the gene intron 3 acceptor site or a novel acceptor site generated by the target duplication caused by transposition. One of them, referred as mRNA type 1, encodes a truncated protein that could be predictably non-functional. In mR…

Meanings of travel in the musical discourse of blues: Roosevelt “Grey Ghost” Williams and “Lonesome Traveller”

2016

Acidification processes in a peritidal carbonate succession across the Triassic/Jurassic boundary (Sicily)

2017

For the first time, a correlation between biostratigraphic events and δ13C curve was attempted along an UpperTriassic-Lower Jurassic peritidal limestone succession cropping out in westernmost Sicily. The peritidal carbonates are organized in shallowing upward cycles characterized by subtidal skeletal mudstone to grainstone, intertidal microbial mats and supratidal paleosoils. About 300 meters of this succession covering the Triassic-Jurassic interval were studied in details. On the base of the macro- and microfossil assemblages from the subtidal facies, four informal units have been recognized along the studied section. Unit R1 (at the base, 111 m thick) is dominated by large megalodonts, r…

Logic, Computing and Biology

2015

Logic and Computing are appropriate formal languages for Biology, and we may well be surprised by the strong analogy between software and DNA, and between hardware and the protein machinery of the cell. This chapter examines to what extent any biological entity can be described by an algorithm and, therefore, whether the Turing machine and the halting problem concepts apply. Last of all, I introduce the concepts of recursion and algorithmic complexity, both from the field of computer science, which can help us understand and conceptualise biological complexity.

Inductive inference of recursive functions: Qualitative theory

2005

This survey contains both old and very recent results in non-quantitative aspects of inductive inference of total recursive functions. The survey is not complete. The paper was written to stress some of the main results in selected directions of research performed at the University of Latvia rather than to exhaust all of the obtained results. We concentrated on the more explored areas such as the inference of indices in non-Goedel computable numberings, the inference of minimal Goedel numbers, and the specifics of inference of minimal indices in Kolmogorov numberings.
