Giant amphiphiles encompassing a hydrophilic β-cyclodextrin (βCD) component and a hydrophobic calix[4]arene (CA4) module undergo self-assembly in aqueous media to afford core-shell nanospheres or nanocapsules, depending on the nanoprecipitation protocol, with high docetaxel (DTX) loading capacity. The blank and loaded nanoparticles have been fully characterized by dynamic light scattering (DLS), ζ-potential measurements and cryo-transmission electron microscopy (cryo-TEM). The data are compatible with the distribution of the drug between the nanoparticle core and the shell, where it is probably anchored by inclusion of the DTX aromatic moieties in βCD cavities. Mucoadhesive polymers in pharmaceutical formulations release drugs in mucosal areas. They interact and fix to mucus via molecular interpenetration, etc ., which increase drug bioavailability. Polymers physicochemical properties affect formulation mucoadhesion, rheological behaviour and drug absorption. Hyaluronan (HA) is selected as a mucoadhesive and biodegradable polymer. Geometric, topological and fractal analyses are carried out with program TOPO. Reference calculations are performed with algorithm GEPOL. Procedure TOPO underestimates molecular volume by 0.7%. Interactions of quinine with polyacrylic and poly-L-glutamic acids in aqueous solutions

Abstract Drug-loaded polymers provide an attractive form for controlled drug delivery systems. A proper knowledge of polymer–drug interactions can aid in the designing of polymers for various drug-delivery applications. In this paper we have investigated the interaction of a drug such as quinine, with synthetic macromolecules such as poly(acrylic acid), PAA, and poly(L-glutamic acid), PGA, at pH 7 and 37 °C by fluorescence spectroscopy and viscometry. The analysis of the binding isotherms revealed that the association process is positive cooperative up to a threshold concentration and then it is negative. Computational Study of Nanosized Drug Delivery from Cyclodextrins, Crown Ethers and Hyaluronan in Pharmaceutical Formulations

Abstract The problem in this work is the computational characterization of cyclodextrins, crown ethers and hyaluronan (HA) as hosts of inclusion complexes for nanosized drug delivery vehicles in pharmaceutical formulations. The difficulty is addressed through a computational study of some thermodynamic, geometric and topological properties of the hosts. The calculated properties of oligosaccharides of D-glucopyranoses allow these to act as co-solvents of polyanions in water. In crown ethers, the central channel is computed. α,β-Poly(N-Hydroxyethyl)-DL-Aspartamide Hydrogels as Drug Delivery Devices

α,β-poly(N-hydroxyethyl)-DL-aspartamide (PHEA) was exposed to gamma radiation to obtain micromatrices able to swell in an aqueous medium. Crosslinked PHEA was loaded with an anti-inflammatory drug, 4-biphenylacetic acid (BPAA) and the drug dispersion in the network was investigated by X-ray analysis. The BPAA loaded PHEA microparticles were also characterized by dimensional analysis, which showed the presence of quasispherical shapes. The drug release from PHEA hydrogel was studied in vitro in a pH 1.1 (simulated gastric juice) and in a pH 7.4 buffer solution, respectively. Attaching targeting ligands on the surface of self-assembled drug delivery systems is the key request for a controlled transport of the drug to a desired location. Reactive nanorods based on activated esterpolymers: a versatile template-assisted approach for the fabrication of functional nanorods

A new route for the fabrication of polymeric nanorods with functional moieties via post-modification of reactive nanorods is described. To this end reactive nanorods with a homogenous and narrow size distribution were fabricated by utilizing an anodic aluminium oxide (AAO) template-assisted approach.
