Application of ultrasound as clean technology for extraction of specialized metabolites from stinging nettle (Urtica dioica L.)
2022
Nettle is a highly valued medicinal plant that is still largely neglected, both in terms of nutrition and use for pharmacological purposes. Tinctures, i.e., alcoholic extracts, are becoming increasingly popular nettle products, mainly because they allow better availability of phytochemicals and their stability over a longer period of time. The production of alcoholic extracts is a chemically demanding process that is still usually carried out using conventional techniques, which have numerous drawbacks. The use of green technologies such as ultrasound-assisted extraction (UAE), which is characterized by high efficiency of phytochemical extraction, shorter treatment time, and a much lower en…
Synthesis and antibacterial activities of cadiolides A, B and C and analogues
2015
International audience; The one-pot multicomponent synthesis of natural butenolides named cadiolides A, B, C and analogues has been realized. The antibacterial structure activity relationship shows that the presence of phenolic hydroxyl groups and the number and position of bromine atoms on the different aromatic rings are important features for antibacterial activity, besides it was demonstrated the tolerance of both benzene and furan ring at position 3 of the butenolide nucleus. Furthermore, none of the most relevant antibacterial compounds showed any cytotoxicity in freshly isolated human neutrophils.
Modulation of adrenergic responses of human vas deferens by K+ channel inhibitors
2010
The results suggest that charybdotoxin-sensitive, but iberiotoxin-insensitive, K+ channels are activated by stimulation with norepinephrine and electrical field stimulation to counteract the adrenergic-induced contractions of human vas deferens. Thus, inhibition of these channels increases significantly the contraction, an effect that appears to be mediated by an increase in Ca2+ entry through L-type voltage-dependent Ca2+ channels.
Histone post-translational modifications as potential therapeutic targets for pain management
2021
Effective pharmacological management of pain associated with tissue pathology is an unmet medical need. Transcriptional modifications in nociceptive pathways are pivotal for the development and the maintenance of pain associated with tissue damage. Accumulating evidence has shown the importance of the epigenetic control of transcription in nociceptive pathways via histone post-translational modifications (PTMs). Hence, histone PTMs could be targets for novel effective analgesics. Here, we discuss the current understanding of histone PTMs in the modulation of gene expression affecting nociception and pain phenotypes following tissue injury. We also provide a critical view of the translationa…
Foxp3 Silencing with Antisense Oligonucleotide Improves Immunogenicity of an Adjuvanted Recombinant Vaccine against Sporothrix schenckii
2021
Made available in DSpace on 2021-06-25T10:56:14Z (GMT). No. of bitstreams: 0 Previous issue date: 2021-04-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Background: In recent years, there has been great interest in developing molecular adjuvants based on antisense oligonucleotides (ASOs) targeting immunosuppressor pathways with inhibitory effects on regulatory T cells (Tregs) to improve immunogenicity and vaccine efficacy. We aim to evaluate the immunostimulating effect of 2′OMe phosphorothioated Foxp3-targeted ASO in an antifungal adjuvanted recombinant vaccine. Methods: The uptake kinetics of Foxp3 ASO, its cyto-toxicity and its ability to deplete Tregs were evaluated in…
Prenatal diazepam exposure functionally alters the GABA(A) receptor that modulates [3H]noradrenaline release from rat hippocampal synaptosomes.
2002
In rats, exposure to diazepam (DZ) during the last week of gestation is associated with behavioral alterations (in some cases sexually dimorphic) that appear when the animals reach adulthood. This study was conducted to evaluate the effects of prenatal DZ exposure on the function of the gamma-aminobutyric (GABA)(A) receptor complex. The method used - perfusion of rat hippocampal nerve terminals labeled with [3H]noradrenaline (NA) - allowed us to evaluate the effects of DZ on a specific native GABA(A) receptor subtype which is located on hippocampal noradrenergic nerve endings and mediates the release of NA. Muscimol stimulated synaptosomal release of [3H]NA in a concentration-dependent mann…
ASSESSMENT INTEGRATO NELLA DIPENDENZA DAL GIOCO D’AZZARDO: DALLA PSICODIAGNOSI AL NEUROIMAGING
GAMBLING: SPREADING AMONG STUDENTS IN PALERMO
2017
Adolescents represent a particularly high-risk group and they are also vulnerable to the development of gambling related problems. Thanks to technological advances in gaming industry (slot machines, video lottery terminals, casino-types games) new form of gambling are continuously appearing. With the purpose of having an idea about the spreading of gambling among the students of Palermo, a survey has been conducted through administration of questionnaires during the period from January to May 2017: this study involved 1622 students (686 boys and 936 girls) between the age of 14 and 19. In this survey it has been demonstrated that only 15% of students have never played; 36% declare they have…
Biological characterization of laryngeal squamous-cell carcinoma
1996
The traditional prognostic factors, including stage of disease and tumour grade, have shown a limited prognostic significance and an inability to predict clinical response to specific treatment in patients with laryngeal squamous-cell carcinoma. More recent data suggest that cell kinetics indices, DNA-ploidy, lysosomal cysteine proteinase expression and genetic changes of both tumour suppressor genes and proto-oncogenes may be considered as reliable and reproducible indicators of biological aggressiveness in these patients. Moreover, the frequency of different genetic alterations suggests that several pathways are involved in the genesis of these neoplasias and, in particular, it is very pr…
