A membrane-destabilizing peptide in capsid protein L2 is required for egress of papillomavirus genomes from endosomes

ABSTRACT Papillomaviruses are internalized via clathrin-dependent endocytosis. However, the mechanism by which viral genomes pass endosomal membranes has not been elucidated. In this report we show that the minor capsid protein L2 is required for egress of viral genomes from endosomes but not for initial uptake and uncoating and that a 23-amino-acid peptide at the C terminus of L2 is necessary for this function. Pseudogenomes encapsidated by L1 and L2 lacking this peptide accumulated in vesicular compartments similar to that observed with L1-only viral particles, and these mutant pseudoviruses were noninfectious. Pyolysin (PLO) belongs to the homologous family of the cholesterol- dependent cytolysins (CDCs), which bind to cell membranes containing cholesterol to form oligomeric pores of large size. The CDC monomer structure consists of 4 domains. Among these, the C-terminal domain 4 has been implicated in membrane binding of the monomer, while the subsequent processes of oligomerization and membrane insertion have primarily been assigned to other domains of the molecule. Recombinantly expressed or proteolytic fragments that span domain 4 of the CDCs streptolysin O and perfringolysin O bind to membranes but fail to oligomerize, and they inhibit the activity of the respective wild-type toxins. Inclusion ratio based estimator for the mean length of the boolean line segment model with an application to nanocrystalline cellulose

A novel estimator for estimating the mean length of fibres is proposed for censored data observed in square shaped windows. Instead of observing the fibre lengths, we observe the ratio between the intensity estimates of minus-sampling and plus-sampling. It is well-known that both intensity estimators are biased. In the current work, we derive the ratio of these biases as a function of the mean length assuming a Boolean line segment model with exponentially distributed lengths and uniformly distributed directions. Monitoring glacier elevation and volume changes with digital photogrammetry and GIS at Gepatschferner glacier, Austria

Digital elevation models of Gepatschferner in Northern Tyrol, Austria were obtained with digital photogrammetry from high altitude stereo photo pairs and by digitizing an analogue topographic glacier map, for 1990 and 1971, respectively. A difference map was calculated to identify regions of glacier elevation increases and decreases corresponding to glacier mass gain and loss. While the glacier tongue below 2600 m showed mainly increases in glacier surface elevation as compared to 1971, the remaining glacier thickness experienced both increases and decreases. Histidine residues near the N terminus of staphylococcal alpha-toxin as reporters of regions that are critical for oligomerization and pore formation.

Chemical modification of histidine residues in staphylococcal alpha-toxin leads to loss of functional activity. Site-directed mutants of the toxin in which each of the four histidine residues was replaced by several amino acids were therefore produced. The mutant proteins were purified and characterized. Exchange of H-259 or H-144 was sometimes tolerated without reduction in hemolytic activity. These histidine residues are thus not essential for toxin function. Exchange of H-35 and H-48, however, had marked effects. H-35 mutant toxins bound with high affinity to rabbit erythrocytes but displayed faulty oligomerization and were unable to form pores. The imperial cult in Late Antiquity (4th-6th cent. AD) can be interpreted as a significant juridical fossile, which conveys the inner contradictions of the so called “era of Constantine”. An epigraphic and prosopographic analysis of the high priests of the imperial cult (flamines, sacerdotes and coronati in the West, ἀρχιερεῖς in the East) would point out the lasting importance of these dignitaries between the ages of Diocletian and Justinian, especially in the case of provincial high priests, who were in the same time presidents of federal assemblies (concilia, κοινά) and played a primary role in the “religious reforms” planned by Maximinus Daia and Julian the Apostate.

Ir uzskatīts, ka TN etioloģijas pamatā ir hroniskais neirovaskulārais kontakts (t.s. neirovaskulārais konflikts) starp asinsvadu un nervu. Rezultātā asinsvads spiež uz nervu, veidojas nervu mielīna apvalka bojājums, pastāvīgs nervu šķiedru nociceptīvo receptoru kairinājums, kas izpaužas ar sāpēm.
