Evaluation of some growth regulator effects on encapsulated in vitro-derived microcuttings of three Italian Ficus carica L. genotypes.

In this study, the encapsulation technology based on the calcium alginate coating was applied to some Ficus carica L. genotypes. Uninodal microcuttings (3-4 mm long), excised from in vitro proliferating shoots of three Italian cultivars (‘Bifera’, ‘Palazzo’ and ‘Catalanisca’), were employed. The influence of three different plant growth regulators (PGRs): 6-benzylaminopurine (BAP), meta-topolin (MT) and zeatine (ZEA), added to the artificial endosperm, were evaluated. Particularly, the viability, regrowth and conversion parameters of the synthetic seeds were registered after 60 days from the sowing on a hormone-free medium. Results showed that the cultivars ‘Catalanisca’ and ‘Palazzo’ showe…

The effects of meta-Topolin and benzyladenine on in vitro organogenesis from epicotyl cuttings of Troyer citrange (Citrus sinensis[L.] Osbeck ×Poncirus trifoliata[L.] Raf.)

Citrus tristeza virus (CTV), worldwide considered as one of the worst disease affecting Citrus plants, produced great changes in citriculture, mainly because it made mandatory the turnover of the sensitive sour orange rootstock with virus resistant ones. Among them, Troyer citrange has become one of the most employed rootstocks, due to its CTV tolerance and to the positive characteristics induced to the scion fruit production. Unfortunately, in Italy, there is a scarcity of seed mother plants. Micropropagation can be a solution for this problem. Among the in vitro techniques allowing the production of good quality true-to-type plants, the regeneration from seedling epicotyl cuttings general…

Cryopreservation of Italian cultivars of hazelnut

The present study was performed to optimise the cryopreservation protocol in Italian cultivars of Corylus avellana L. Lateral buds excised from in vitro grown plantlets of ‘Tonda Gentile Romana’ (TGR) and ‘Montebello’ (MB) were cryopreserved by the encapsulation-dehydration technique. The effect of dehydration of beads with sucrose (0.5, 0.75 or 1 M) for 1 or 3 days, of desiccation with silica gel for 6 or 8 h and of application of two cytokinins (N6-benzyladenine or meta-Topolin) in the regrowth phase was evaluated. The highest regrowth was obtained dehydrating alginate beads with 0.75 M sucrose applied for 1 day and with desiccation for 8 h with silica gel in both cultivars. On the other …
