COMPARATIVE STUDY ON THE ELECTROPHYSIOLOGICAL EFFECTS OF MONENSIN ON ATRIAL AND VENTRICULAR MUSCLE PREPARATIONS OF GUINEA-PIG HEART
The effects of monensin (10 µmol/L) alone and in presence of verapamil (3 µmol/L) or ouabuin (1 µmol/L) or glibenclamide (10 µmol/L) or BaCl2 separately were studied on the electrophysiological properties represented by force of contraction, duration of action potential, and membrane resting potential of isolated atrial and ventricular muscle of guinea –pig heart. Monensin induced a transient increase in the force of contraction of atrial and ventricular muscles. It caused a decrease in action potential duration of papillary muscle, this effect was reversed by glibenclamide. Monensin increased the action potential duration of atrial muscle in absence or presence of glibenclamide. In presenc…
MECHANISMS OF TENSION DEVELOPMENT INDUCED BY MONENSIN IN GUINEA-PIG AORTA : EFFECTS OF VERAPAMIL, OUABAIN, PRAZOSIN AND GLIBENCLAMIDE
The influence of monensin (10 µmol/L) alone and in the presence of verapamil (3 µmol/L) or ouabain (1 mmol) and prazosin (1 µmol/L) or glibenclamide (10 µmol/L) were studied on the muscle tension of guinea pig aorta. Changes in tissue sodium, potassium and calcium ion contents of the aortic muscle produced by monensin in the presence of prazosin were evaluated. Monensin in normal Tyrode's solution containing prazosin caused an increase in the resting tension followed by a decrease and returned to normal values. Verapamil reduced the muscle tension induced by monensin. After ouabain, monensin induced sustained increase in the resting tension. Glibenclamide partially reversed the relaxant pha…
MONENSIN ENHANCES DIGOXIN-INDUCED ARRHYTHMIAS IN GUINEA-PIGS
Effects of pretreatment with monensin (150 ug/kg), atenolol (0.3 mg/kg), atenolol plus monensin, verapamil (0.38 mg/kg), verapamil plus monensin, glibenclamide(0.38 mg/kg) and glibenclamide plus monensin on the dose of digoxin required to induce premature ventricular contractions (PVCS) in anaesthetized guinea-pigs were studied. Monensin reduced while atenolol increased the dose of digoxin required to produce PVCS. Atenolol plus monensin increased the dose of digoxin required to produce PVCS in presence of monensin alone. Verapamil reduces the arrhythmogenic effect of monensin on digoxin. Glibenclamide antagonises the effect of monensin on digoxin induced PVCS. From the present data it coul…
