Possible relationship between micronucleated and binucleated cells induced by cisplatin in cultured CHO cells

Abstract Chinese hamster ovary (CHO) cells were treated with a single dose (10 μg/ml) of cis-diammino-dichloroplatinum (II) (cisplatin) for 1 h and the effect of the drug on the kinetics of proliferation of the cultures was studied. It was found that the drug produces a delay in the proliferation rates of the treated cultures. The induction of micronuclei and binucleated cells (BC) at different times after treatment have also been studied, and the ability of these cells to undergo DNA synthesis (measured as the ability to incorporate [3H]thymidine) is shown. It was found that cisplatin induced a particular type of BC that contains one or more micronuclei rather than a pure population of BC.…

Effect of chemical enhancers on the in vitro percutaneous absorption of sumatriptan succinate

The effects of percutaneous enhancers on the transdermal absorption of sumatriptan succinate were investigated by in vitro permeation studies. Pretreatment of porcine skin with ethanol (vehicle), polyethylene glycol 600, Span 20, oleic acid, R-(+)-limonene, alpha-bisabolol and 1,8-cineole (at 5% in ethanol, w/w) produced in all cases an increase in the flux of sumatriptan. The amount of sumatriptan retained in the skin was also determined. Ethanol has showed a low but significant increment on the drug transdermal flux. High GSH content associates with high metastatic activity in B16-F10 melanoma cells cultured to low density (LD B16M). GSH homeostasis was investigated in LD B16M cells that survive after adhesion to the hepatic sinusoidal endothelium (HSE). Invasive B16M (iB16M) cells were isolated using anti-Met-72 monoclonal antibodies and flow cytometry-coupled cell sorting. HSE-derived NO and H(2)O(2) caused GSH depletion and a decrease in gamma-glutamylcysteine synthetase activity in iB16M cells. B16 melanoma (B16M) cells with high glutathione (GSH) content show rapid proliferation in vitro and high metastatic activity in the liver in vivo. gamma-Glutamyl transpeptidase (GGT)-mediated extracellular GSH cleavage and intracellular GSH synthesis were studied in vitro in B16M cells with high (F10) and low (F1) metastatic potential. High-performance liquid chromatographic determination of sumatriptan after in vitro transdermal diffusion studies.

A simple, accurate, precise and rapid HPLC method with UV detection has been validated in order to determine the in vitro transdermal absorption of sumatriptan succinate. The HPLC method is a modification of that described by Nozal et al. [M.J. Nozal, J.L. Bernal, L. Toribio, M.T. Martin, F.J. Diez, J. Pharm. Biomed. Anal. 30 (2002) 285-291]. Separation was carried out on a 250 mm Kromasil C18 column at room temperature. The detector response, at 282.7 nm, was found to be linear in a concentration range between 0.145 and 145 microM. The limit of detection (LOD) was 0.019 microM and the limit of quantification (LOQ) was 0.145 microM.

Abstract The mutagenicity and cytotoxicity of cis -diamminedichloroplatinum (II) (cisplatin) at doses of 5, 10 and 20 μg/ml in Chinese hamster ovary (CHO) cells have been examined. A morphological characterization of several cell types induced by cisplatin was carried out. The frequencies of both cells with micronuclei and binucleate cells as a time-dependent parameter have also been studied. Whilst the number of cells with micronuclei was found to decrease with time, the number of binucleate cells increased.
