“Click” on PLGA-PEG and hyaluronic acid: Gaining access to anti-leishmanial pentamidine bioconjugates

Pentamidine (Pent), an antiparasitic drug used for the treatment of visceral leishmaniasis, has been modified with terminal azide groups and conjugated to two different polymer backbones (PLGA-PEG [PP] copolymer and hyaluronic acid [HA]) armed with alkyne end-groups. The conjugation has been performed by Copper Catalyzed Azido Alkyne Cycloaddition (CuAAC) using CuSO4 /sodium ascorbate as metal source. The novel PP-Pent and HA-Pent bioconjugates are proposed, respectively, as non-targeted and targeted drug delivery systems against Leishmania infections. Amphiphilic cyclodextrin (ACyD) provides water-soluble and adaptable nanovectors by modulating the balance between the hydrophobic and hydrophilic chains at both CyD sides. This work aimed to design nanoassemblies based on nonionic and hydrophilic ACyD (SC6OH) for the delivery of a poor-water-soluble organotin(IV)-porphyrin derivative [(Bu3Sn)4TPPS] to melanoma cancer cells. Abstract Highly de-bundled multiwalled carbon nanotube (MWCNT) polymer nanohybrids were obtained via ionic assembling of a novel POSS-polyimidazolium-ionic liquid (POSS = polyhedral oligomeric silsesquioxanes) with oxidized MWCNTs. The synthesis, accomplished by a simple bilayer chloroform/water metathetical reaction process, was promoted by the high surfactant properties of POSS-imidazolium-polyionic liquid, which assisted the effective de-bundling, dispersion and purification of the as prepared POSS@MWCNT polymers in organic media, via a surface coating mechanism. Leishmaniasis is still a serious public health problem worldwide, especially in tropical areas where this infectious disease is endemic. The most severe form of the disease (i.e. visceral) can claim victims if left untreated and the few accessible drugs have several drawbacks including major side effects and parenteral administration.
