Metallothionein genes in sea urchin embryos and their induction by cadmium

Metallothioneins (MTs) are proteins that play a major role in metal homeostasis and as a reservoir. The MT gene/protein systems of sea urchins are an invaluable model for the study of gene expression regulation and MT isoform-specific functionality of these proteins. We isolated five paralogous MT isogenes and we studied their expression in the sea urchin embryo Paracentrotus lividus. The Cadmium-dependent transcriptional activation of the five isogenes was assessed using quantitative Real Time PCR. Metallothioneins (MT) are small and cysteine-rich proteins that bind metal ions such as zinc, copper, cadmium, and nickel1. Several functions were proposed for these peptides, ranging from toxic metal protection to physiological metal homeostasis, free radical scavenging, oxidative stress protection, antiapoptotic defense, control of the redox status of the cell and also roles during development. Hsp56 protein and mRNA distribution in normal and stressed P. lividus embryos

It was previously demonstrated that Paracentrotus lividus Hsp56 mitochondrial chaperonin is con- stitutively expressed during development, that it increases after heat-shock and cadmium treatment, and that it has a specific territorial distribution, both in normal and heat-shocked embryos, as shown by immunolocalization experiments. In this work, we analyzed by Western blot the territorial distribution of the protein in plutei exposed to heat-shock or sublethal cadmium concentrations, and we found that Hsp56 increases in both ectodermal and en- dodermal cells. Cellular nucleic acid binding proteins (CNBPs) represent a highly conserved protein family among vertebrates; they harbour seven tandem zinc finger repeats CCHC type and have been described as transcriptional and translational regulators.
