Precision wormlike nanoadjuvant governs potency of vaccination

It remains unclear how the precise length of one-dimensional nanovehicles influences the characters of vaccination. Here, a unimolecular nanovehicle with tailored size and aspect ratio (AR) is applied to deliver CpG oligodeoxynucleotide, a Toll-like receptor (TLR) 9 agonist, as an adjuvant of recombinant hepatitis B virus surface antigen (rHBsAg), for treating chronic hepatitis B (CHB). Cationic nanovehicles with fixed width (ca. 45 nm) but varied length (46 nm-180 nm), AR from 1 to 4, are prepared through controlled polymerization and are loaded with CpG by electrostatic interaction. We reveal that the nanoadjuvant with AR = 2 shows the highest retention in proximal lymph nodes. Importantl…

Composite Thin Film by Hydrogen-Bonding Assembly of Polymer Brush and Poly(vinylpyrrolidone)

Based on hydrogen-bonding layer-by-layer (LBL) assembly in aqueous solution, poly(vinylpyrrolidone) (PVPON) and a spherical polymer brush with a poly(methylsilsesquioxane) (PSQ) core and poly(acrylic acid) (PAA) hair chains were used to fabricate composite multilayer thin films. Hydrogen bonding as the driving force was confirmed by FT-IR spectrometry. A simple method (Filmetric F20) was introduced to determine the thickness and refractive index of the films. The film thickness was found to be a linear function of the number of bilayers. The average increase in thickness per bilayer is 28.3 nm. The film morphology was characterized with scanning electron microscopy and atomic force microsco…

Functionalization of Crosslinked Vesicles by Co-Self-Assembly of a Gelable Diblock Copolymer and Mercaptosilane

Herein, a convenient and general method to simultaneously fix and functionalize polymeric vesicles with sulphydryl groups by the co-self-assembly of poly(ethylene oxide)-block-poly[3-(triethoxysilyl)propyl methacrylate] (PEO-b-PTESPMA) and 3-mercaptopropyltrialkoxysilane in an aqueous solution is reported. The presence of sulphydryl groups across the vesicle membrane has been confirmed by using an energy-filtered technique during TEM analysis and by capturing Au nanoparticles.

Influence of Hair Density and Hair Length on Interparticle Interactions of Spherical Polymer Brushes in a Homopolymer Matrix

The dynamics of hairy spherical nanoparticles in a melt of linear polymer chains has been investigated by mechanical spectroscopy as a function of particle topology and concentration. Using a simple free volume approach for the data analysis of the structural relaxation time vs concentration and the well-known hard-sphere result as a reference, a semiquantitative measure for the interparticle interactions, that is particle deformability/softness, and the effective particle size compared to the size of a nonswollen spherical brush has been determined. For these studies, model particles of hairy nanoparticles differing in either hair length or grafting density have been prepared. In contrast …
