Soluble liver antigen (SLA) antibody detection by ELISA and multiplexing technologies

Abstract Objective: Develop and evaluate assays for the detection of antibodies to soluble liver antigen (SLA). SLA, also known as liver/pancreas antibody, was found to be 100% specific for autoimmune hepatitis (AIH) in a recent study of 2000 sera collected from individuals with various disease conditions and healthy individuals (Wies et al., Lancet 2000; 355:1510). Although SLA antibodies occur in only about 30% of patients with autoimmune hepatitis, they are found in some individuals with AIH who are negative for other autoantibodies. Methods: Specimens from patients with autoimmune hepatitis, non-autoimmune liver disease, various autoimmune conditions, as well as specimens from healthy i…

Gluten-Free Diet Reduces Symptoms, Particularly Diarrhea, in Patients With Irritable Bowel Syndrome and Antigliadin IgG

Background & Aims Many patients with irritable bowel syndrome (IBS) perceive that their symptoms are triggered by wheat-containing foods. We assessed symptoms and gastrointestinal transit before and after a gluten-free diet (GFD) in unselected patients with IBS and investigated biomarkers associated with symptoms. Methods We performed a prospective study of 50 patients with IBS (ROME III, all subtypes), with and without serologic reactivity to gluten (antigliadin IgG and IgA), and 25 healthy subjects (controls) at a university hospital in Hamilton, Ontario, Canada, between 2012 and 2016. Gastrointestinal transit, gut symptoms, anxiety, depression, somatization, dietary habits, and microbiot…

Prevalence estimation of celiac disease in the general adult population of Latvia using serology and HLA genotyping

Background: Prevalence estimates for celiac disease (CD) depend on the method used. The role of deamidated gliadin peptide (DGP) and genetic testing in epidemiological studies and diagnostic settings of celiac disease (CD) has still to be established. Objectives: The objective of this article is to assess the prevalence of CD in Latvia by combining serological tests with DQ2.5/ DQ8 testing. Methods: A total of 1444 adults from a randomly selected cross-sectional general population sample were tested by ELISA for tTG IgA, DGP IgA and IgG antibodies (QUANTA Lite, Inova Diagnostics Inc). Samples with tTG IgA 20U were tested for EMA IgA by indirect immunofluorescence assay, and all specimens wi…
