Superresolution imaging of biological nanostructures by spectral precision distance microscopy

2011

For the improved understanding of biological systems on the nanoscale, it is necessary to enhance the resolution of light microscopy in the visible wavelength range beyond the limits of conventional epifluorescence microscopy (optical resolution of about 200 nm laterally, 600 nm axially). Recently, various far-field methods have been developed allowing a substantial increase of resolution ("superresolution microscopy", or "lightoptical nanoscopy"). This opens an avenue to 'nano-image' intact and even living cells, as well as other biostructures like viruses, down to the molecular detail. Thus, it is possible to combine light optical spatial nanoscale information with ultrastructure analyses…

Structure and reactivity of a mononuclear gold(II) complex.

2017

Mononuclear gold(II) complexes are very rare labile species. Transient gold(II) species have been suggested in homogeneous catalysis and in medical applications, but their geometric and electronic structures have remained essentially unexplored: even fundamental data, such as the ionic radius of gold(II), are unknown. Now, an unprecedentedly stable neutral gold(II) complex of a porphyrin derivative has been isolated, and its structural and spectroscopic features determined. The gold atom adopts a 2+2 coordination mode in between those of gold(III) (four-coordinate square planar) and gold(I) (two-coordinate linear), owing to a second-order Jahn–Teller distortion enabled by the relativistical…

Structure‐Activity Relationships of Benzamides and Isoindolines Designed as SARS‐CoV Protease Inhibitors Effective against SARS‐CoV‐2

2020

Abstract Inhibition of coronavirus (CoV)‐encoded papain‐like cysteine proteases (PLpro) represents an attractive strategy to treat infections by these important human pathogens. Herein we report on structure‐activity relationships (SAR) of the noncovalent active‐site directed inhibitor (R)‐5‐amino‐2‐methyl‐N‐(1‐(naphthalen‐1‐yl)ethyl) benzamide (2 b), which is known to bind into the S3 and S4 pockets of the SARS‐CoV PLpro. Moreover, we report the discovery of isoindolines as a new class of potent PLpro inhibitors. The studies also provide a deeper understanding of the binding modes of this inhibitor class. Importantly, the inhibitors were also confirmed to inhibit SARS‐CoV‐2 replication in …

Entering the Nano-Cosmos of the Cell by Means of Spatial Position Determination Microscopy (SPDM): Implications for Medical Diagnostics and Radiation…

2013

During the last 20 years fluorescence light microscopy has made an enormous progress towards fluorescence nanoscopy in order to elucidate the nanostructural organization of cellular machineries beyond classical limits of resolution in light microscopy. One of these novel techniques is Spatial Position Determination Microscopy (SPDM), an approach of molecular localization microscopy based on the application of specific fluorescence labelling of cellular structures by means of dyes that undergo reversible photobleaching resulting in blinking effects during image acquisition. This blinking allows spectral separation of individual molecules and thus precise localization and distances measuremen…

Imprinting the complex dielectric permittivity of liquids into the spintronic terahertz emission

2021

We report an approach in time-domain terahertz (THz) spectroscopy for measuring the dielectric response of liquids based on inherent properties of spintronic THz emitters (STEs). The THz electric field radiated from the STE is inversely proportional to the sum of the complex refractive indices of the media surrounding the thin metallic stack of the STE and the stack's conductivity. We demonstrate that by bringing a liquid in contact with the emitter, its complex refractive index and accordingly its dielectric response are imprinted into the radiated electromagnetic field from the emitter. We use water as the test liquid and ascertain its dielectric loss and permittivity in the range of ∼0.…

Interfering with Host Proteases in SARS-CoV-2 Entry as a Promising Therapeutic Strategy

2020

Abstract: Due to its fast international spread and substantial mortality, the coronavirus disease COVID-19 evolved to a global threat. Since there is currently no causative drug against this viral infection available, science is striving for new drugs and other approaches to treat the new disease. Studies have shown that the cell entry of coronaviruses into host cells takes place through the binding of the viral spike (S) protein to cell receptors. Priming of the S protein occurs via hydrolysis by different host proteases. The inhibition of these proteases could impair the processing of the S protein, thereby affecting the interaction with the host-cell receptors and preventing virus cell …
