Quantification of the Detrimental Effect of a Single Primer-Template Mismatch by Real-Time PCR Using the 16S rRNA Gene as an Example

ABSTRACT We investigated the effects of internal primer-template mismatches on the efficiency of PCR amplification using the 16S rRNA gene as the model template DNA. We observed that the presence of a single mismatch in the second half of the primer extension sequence can result in an underestimation of up to 1,000-fold of the gene copy number, depending on the primer and position of the mismatch.

Most examples of intercontinental dispersal events after the Miocene contact between Africa and Asia involve mammal lineages. Among amphibians, a number of probably related groups are known from both continents, but their phylogenies are so far largely unresolved. To test the hypothesis of Miocene dispersal against a Mesozoic vicariance scenario in the context of Gondwana fragmentation, we analyzed fragments of the mitochondrial 16S rRNA gene (572 bp) in 40 specimens of 34 species of the anuran family Ranidae. Abstract In 1648, an uprising broke out in Ukraine that belonged to Poland at the time. The war was not successful for the Polish-Lithuanian Commonwealth. The Cossacks and Tartars, who were helping them during the war, surrounded Zbaraż, about whose defense everyone knew in the country. In a previous article [Oggerin M., Arahal, D. R., Rubio, V. & Marin, I. (2009). Int J Syst Evol Microbiol 59, 2323–2328], it has been shown that strain Beijerinckia fluminensis UQM 1685T and its derived equivalent B. fluminensis CIP 106281T do not conform to the description of the type strain of Beijerinckia fluminensis Döbereiner and Ruschel 1958. Indeed, both strains were identified as members of the species Rhizobium radiobacter and exhibited marked phenotypic and genotypic differences with members of the genus Beijerinckia. Mapping and determinism of soil microbial community distribution across an agricultural landscape.

The effects of increasing atmospheric CO2 on ocean ecosystems are a major environmental concern, as rapid shoaling of the carbonate saturation horizon is exposing vast areas of marine sediments to corrosive waters worldwide. Natural CO2 gradients off Vulcano, Italy, have revealed profound ecosystem changes along rocky shore habitats as carbonate saturation levels decrease, but no investigations have yet been made of the sedimentary habitat.
