Aneuploidy, the alteration of the normal number of chromosomes, is found in most of the human solid tumors and correlated with to defects in the process of chromosome segregation (1). It was also suggested that the alteration of the 5-methylcytosine (5-mC) pattern in the chromosome pericentromeric region, generated to aneuploid cells (2, 3). To investigate the relationship between hypomethylation and whole chromosome aneuploidy, we treated HCT-116 cells, a near diploid line, with the demethylating agent 5-aza- 2'-deoxycytidine (DAC). Recent studies suggest that Retinoblastoma tumor suppressor (RB) plays important roles in the prevention of chromosomal instability by regulating genes that control cell cycle progression and mitotic events. We investigated the effects of stable post-transcriptional silencing of RB in primary human fibroblasts (IMR90) and in near-diploid colon cancer cells (HCT116) focusing on chromosome missegregation mechanisms. Stem cells are a minor population of mostly resting cells defined by their long life, high clonogenicity, self-replicating potential, plasticity, and drug resistance (Finn, 2008). BACKGROUND: Changes in chromosome number or structure as well as supernumerary centrosomes and multipolar mitoses are commonly observed in human tumors. Thus, centrosome amplification and mitotic checkpoint dysfunctions are believed possible causes of chromosomal instability.
