The histone deacetylase inhibitor ITF2357 targets oncogenic BRAF in human melanoma cells
2016
ITF2357 (Givinostat) is a potent antineoplastic histone deacetylase inhibitor which is currently used in clinical trials for leukemias and myelomas and in the therapy for systemic juvenile idiopathic arthritis. Here evidence is provided that ITF2357 reduces the viability of human melanoma SK-Mel28 cells thereby inducing cell death. This compound was more efficacious than SAHA, another well known HDAC inhibitor belonging to the same class of hydroxamic acids. Moreover, we demonstrated for the first time that ITF2357 determines in SK-Mel28 cells a remarkable reduction in the level of oncogenic B-Raf, the product of the BRAF V600E mutated gene in melanoma. Western blot analysis showed that the…

White light-emitting diodes (WLEDs) are big news in the field of lighting, however, current production processes are still very expensive or based on unsustainable inorganic metals such as inorganic phosphorus. The EU-funded ARTIBLED project aims to produce low-cost and high-efficiency Bio-hybrid light-emitting diodes (Bio-HLEDs). This can be achieved using artificially synthesized fluorescent proteins linked in biological scaffolds like the packaging to obtain LED for lighting applications containing a biogenic phosphor. This study aims to optimize the protein scaffold CTPR10 production process to obtain a high number of scaffolds with a good purity level for Bio-HLEDs construction. Differ…
Evaluation of the in vitro and in vivo antineoplastic effects of Parthenolide on MDA-MB231 breast cancer cells
2012
Triple-negative breast cancer refers to an aggressive subtype of breast cancer in which the tumor cells lack receptors for estrogen, progesterone and the HER2 protein on their surfaces. This type of breast cancer does not respond to treatments such as hormone therapy, like tamoxifen and aromatase inhibitors, or drugs that target HER2, like Herceptin. It is important, therefore, the identification of new selective drugs for the treatment of these tumors. Parthenolide (PN), a sesquiterpene lactone extracted from the medical plant Tanacetum parthenium, exerts anticancer activity on several tumor cell lines in culture, acting through diverse molecular mechanisms. Our previous studies have shown…

FORENSIC INVESTIGATION OF KERATIN MATRIX: DEVELOPMENT OF A METHOD BY TURBOFLOW™ HPLC-MS/MS FOR CANNABINOIDS QUANTITATIVE ANALYSIS
2016
The products of cannabis are the most frequently used illicit drugs of abuse detected in workplace drug-testing programs or in cases of driving under the influence of drugs. For this reason, there is a great need for sensitive and specific methods which can confirm the chronic exposure and which take account of the cut-off values that have been proposed in Italy by the GTFI Guidelines (Group of Italian Forensic Toxicologists). The method currently more appropriate for quantification of Δ9-Tetrahydrocannabinol (THC) and its major metabolite 11-nor-9-Carboxy-Δ9-THC (THCA) appears to be one that involves HPLC-MS/MS and keratin matrix. The purpose of this work, performed at the Mass Spectrometr…
