Nanocarriers to targeted terapy: a goal for biomedical applications.

2011

The reasons for the growing interest in biomaterials for application in the biomedical field are related to need to find alternative and more effective than conventional procedures used in drug treatment of several diseases. The traditional methods of drug delivery has limitations due to the drug’s toxicity in the body. The development of nanocarriers, which can be functionalized and loaded with drugs or genetic material and can be released into the body with a controlled mechanism, was a real goal in the field of biomedical applications. Synthetic hydrogels have found many applications especially in tissue-engineering scaffolds, as carriers for implantable and drug delivery devices. Hydrog…

Melanoma cells release extracellular vesicle which contain H1° linker histone as well as RNA-binding proteins which bind to the H1° mRNA

2015

We previously demonstrated that G26/24 oligodendroglioma cells release EVs that contain proteins, such as FasL and TRAIL, which induce apoptosis in rat cortical neurons [1] and astrocytes [2]. We also reported that cancer cells use EVs for transferring, into the environment [3], proteins such as extracellular matrix remodelling proteases [4], and H1°, a differentiation-specific histone [5]. In particular, by releasing H1°, cells could escape differentiation cues [5]. To verify the role of EVs in releasing specific proteins and mRNAs, in this study we used as a model A375 melanoma cells. METHODS EVs were purified from cell culture media as previously reported [1, 2]. T1 RNase-protection assa…

Role of sparc and MIR-29B1 in molecular effects induced by win in osteosarcoma MG63 cells

2014

SPARC (Secreted protein acidic and rich in cysteine) is considered as a prototype of matricellular protein due to its structure and the function that it displays in regulating cell/extracellular microenvironment interactions during development and in response to injury. Earlier studies underlined pleiotropic effects of intracellular SPARC on cancer growth and, in some cancer cell lines, identified it as a tumor suppressor protein. Objective This study aimed to evaluate the role of SPARC and its related miRNA in the molecular effects induced by the cannabinoid WIN in osteosarcoma MG63 cells. In these cells WIN is not able to induce cell death but sensitizes cells to TRAIL-mediated apoptotic …
