In vitro models of BBB: a tool for the analysis of cell to cell communication in the brain

2008

Many researchers have been trying to set in vitro models of the blood-brain barrier (BBB) aimed at analyzing, in simplified terms, the molecular mechanisms responsible for formation, maintenance and functioning of the BBB, as well as the capability of specific drugs and pro-drugs to cross BBB. We did it, starting with a simpler system of co-culture that allowed us to analyze the effects of neurons on differentiation of brain capillary endothelial cells (RBE4.B cells) in culture, and setting then a more complex model, that includes three cell types (endothelial cells, neurons and astrocytes). The reciprocal geometrical organization of brain cells in this model system is similar to the one ob…

Effect of extracellular vesicles from CSF of multiple sclerosis patients and healthy controls on astrocytes in culture

2017

INTRODUCTION Secretion of extracellular vesicles (EVs) and EV-mediated exchange of molecules among brain cells, under physiological conditions, has been described years ago. At the same time, ability of EVs to transfer proteins and nucleic acids to the surrounding cells, thus modifying their phenotypes, has been clearly demonstrated for tumor cells of different origins, including brain cancers. More recently, involvement of EVs in pathological processes affecting the Central Nervous System (CNS), and their capacity of transferring damaging cargoes have been also suggested. To explore these functions, we exposed primary cultures of astrocytes to EVs isolated from cerebral spinal fluid (CSF) …

Antioxidant activity of cactus pear fruits and bioavailability of betalan components in healthy humans

2008

Consumption of fruit and vegetables is now associated with protection against various pathologies, including cancer and cardiovascular and cerebrovascular diseases, in which oxidative damage is an important etiologic factor. Various compounds from plant food, acting independently of known nutrients and micronutrients, have appeared very important in protecting human health. Most of these components can act as antioxidants or affect redox-dependent cell activities involved both in physiological and pathological events. In this scenery fruits of cactus pear (Opuntia ficus-indica), and the characteristic betalain pigments have recently attracted the attention of researchers. While suggesting m…

Distribution and Function of Gap Junction Coupling in Cortical GABAergic Neurons.

2013

Although gap junctions have been observed in GABAergic interneurons of several brain regions, this chapter focuses on the distribution and functions of gap junctions and connexins in inhibitory interneurons of the cerebral cortex and hippocampus. Evidence for interconnections mediated by electrical synapses is reported for at least eight cerebral cortex interneuron types, classified on the basis of morphology, electrophysiology and molecular markers. The main differences in the organization of these interneuronal networks are summarized in terms of homologous and heterologous electrical coupling and mutual chemical inhibition. The role of connexin36 (Cx36) in forming neuronal electrical syn…

Glycated albumin is correlated to insulin resistance and β-cell secretory function in subjects at risk for developing diabetes

2018

Insulin resistance and β-cell secretory function represent two main issues in the pathogenesis of type 2 diabetes mellitus (T2DM). Conflicting results have been obtained about the association between glycated albumin (GA) and body mass index (BMI), insulin resistance and β-cell function in diabetic patients. Actually, the relationship (if any) between GA and the markers of glucose homeostasis and insulin resistance in subjects at risk of developing diabetes, has not been completely elucidated yet. Two hundred and one patients undergoing to oral glucose tolerance test (OGTT) were enrolled in the study. Routine laboratory tests, including fasting insulin, were performed at enrollment. GA was …
