Algerian Olive Germplasm and Its Relationships with the Central-Western Mediterranean Varieties Contributes to Clarify Cultivated Olive Diversificati…
2021
Olive tree with its main final product, olive oil, is an important element of Mediterranean history, considered the emblematic fruit of a civilization. Despite its wide diffusion and economic and cultural importance, its evolutionary and phylogenetic history is still difficult to clarify. As part of the Mediterranean basin, Algeria was indicated as a secondary diversification center. However, genetic characterization studies from Maghreb area, are currently underrepresented. In this context, we characterized 119 endemic Algerian accessions by using 12 microsatellite markers with the main goal to evaluate the genetic diversity and population structure. In order to provide new insights about …
Phylogenetic Relationship Among Wild and Cultivated Grapevine in Sicily: A Hotspot in the Middle of the Mediterranean Basin
2019
Grapevine (Vitis vinifera ssp. sativa) is a perennial crop especially important for wine and fruit production. The species is highly polymorphic with thousands of different varieties selected by farmers and clonally propagated. However, it is still debated whether grapevine domestication from its wild ancestor (V. vinifera ssp. sylvestris) has been a single event or rather it occurred on multiple occasions during the diffusion of its cultivation across the Mediterranean. Located in the center of the Basin, Sicily is its largest island and has served as a hotspot for all civilizations that have crossed the Mediterranean throughout history. Hundreds of unique grapevine cultivars are still cul…
Parentage Atlas of Italian Grapevine Varieties as Inferred From SNP Genotyping.
2021
The Italian grape germplasm is characterized by a high level of richness in terms of varieties number, with nearly 600 wine grape varieties listed in the Italian National Register of Grapevine Varieties and with a plethora of autochthonous grapes. In the present study an extended SNP genotyping has been carried out on Italian germplasm of cultivated Vitis vinifera subsp. sativa and Vitis hybrids. Several hundred Italian varieties maintained in the repositories of scientific Institutions and about one thousand additional varieties derived from previous studies on European, Southern Italy, Magna Graecia and Georgian germplasm were considered. The large genotyping data obtained were used to ch…
Genetic characterization of an almond germplasm collection and volatilome profiling of raw and roasted kernels
2021
AbstractAlmond is appreciated for its nutraceutical value and for the aromatic profile of the kernels. In this work, an almond collection composed of 96 Sicilian accessions complemented with 10 widely cultivated cultivars was phenotyped for the production of volatile organic compounds using a proton-transfer time-of-flight mass spectrometer and genotyped using the Illumina Infinium®18 K Peach SNP array. The profiling of the aroma was carried out on fresh and roasted kernels enabling the detection of 150 mass peaks. Sixty eight, for the most related with sulfur compounds, furan containing compounds, and aldehydes formed by Strecker degradation, significantly increased during roasting, while …
Characterization of Sicilian rosemary (Rosmarinus officinalis L.) germplasm through a multidisciplinary approach.
2020
In Sicily, small differences exist between wild and cultivated rosemary biotypes; VOCs and genetic profiles may be a useful tool to distinguish them. A germplasm collection of Rosmarinus officinalis L. was harvested from 15 locations in Sicily. Eleven wild and four cultivated populations were collected and, due to the surveyed area covered, they can be considered as a representative panel of Sicilian genetic background of the species. Ex situ plant collection was transferred to the field cultivation in homogeneous conditions for characterizing through a multidisciplinary approach. The study included morphological traits observations (growth habitus, flower color, number and size of leaves, …
Urgent need for preservation of grapevine (Vitis vinifera L. Subsp. vinifera) germplasm from small circum-Sicilian islands as revealed by SSR markers…
2016
Since the last decades grapevine germplasm is undergoing a process of rapid genetic erosion. This process is of particular concern in minor circum-Sicilian islands, because of the sharp reduction of the cultivated surfaces and the shift of their economy from agriculture to tourism. Aiming at valorising and preserving the surviving varieties we collected 185 accessions during several surveys since 2007. Six nuclear microsatellite markers were used for germplasm characterization, yielding 75 different genetic profiles. We found out that most genetic profiles (39) were not listed in national and international grapevine databases, confirming that the Sicilian minor islands represent underexplor…
Factors affecting somatic embryogenesis in eight Italian grapevine cultivars and the genetic stability of embryo-derived regenerants as assessed by m…
2016
Abstract Embryogenic cultures have been used in cryopreservation, genetic transformation, propagation, virus elimination, induced mutagenesis and in many other biotechnological applications, providing excellent opportunities for biotechnology advances in grapevine. Unfortunately the efficiency of somatic embryogenesis (SE) is genotype-dependent in addition to showing interaction with explant type used and the plant growth regulator (PGR) composition. In order to identify the interaction of these parameters in SE, we tested eight wine grapevine cultivars, three explant types (ovary, anther/filament and stigma/style) and four PGR combinations in a statistically designed experiment. The genoty…
High-throughput 18K SNP array to assess genetic variability of the main grapevine cultivars from Sicily
2016
The viticulture of Sicily, for its vocation, is one of the most important and ancient forms in Italy. Autochthonous grapevine cultivars, many of which known throughout the world, have always been cultivated in the island from many centuries. With the aim to preserve this large grapevine diversity, previous studies have already started to assess the genetic variability among the Sicilian cultivars by using morphological and microsatellite markers. In this study, simple sequence repeat (SSR) were utilized to verify the true-to-typeness of a large clone collection (101) belonging to 21 biotypes of the most 10 cultivated Sicilian cultivars. Afterwards, 42 Organization Internationale de la Vigne…
Genetic diversity in a collection of Italian long storage tomato landraces as revealed by SNP markers array
2019
Tomato (Solanum lycopersicum L.) is one of the most important crops worldwide. In this study, we used 7720 genome-wide SNPs to characterize the genetic diversity within a tomato germplasm collection enriched with 64 accessions from southern Italy of the so called "da serbo" type i.e. drought-tolerant and long storage landraces. Notwithstanding the relatively small collection area, 1575 (20.4%) polymorphic SNPs, mostly on Chr11, detected considerable levels of genetic variation. Maximum parsimony analysis of genetic distance revealed four main clusters and clearly separated most "da serbo" landraces from the outgroups. One of the clusters grouped the landraces from the Mount Vesuvius area, t…
