Nonalcoholic fatty liver disease (NAFLD) is strongly associated to the features of metabolic syndrome which can progress to cirrhosis, liver failure and hepatocellular carcinoma. However, the most common cause of mortality in people with NAFLD is not liver-related but stems from atherosclerotic cardiovascular disease (CVD). The prevalence of NAFLD is on the rise, mainly as a consequence of its close association with two major worldwide epidemics, obesity and type 2 diabetes (T2D). Confidence-based Somatic Mutation Evaluation and Prioritization

Next generation sequencing (NGS) has enabled high throughput discovery of somatic mutations. Detection depends on experimental design, lab platforms, parameters and analysis algorithms. However, NGS-based somatic mutation detection is prone to erroneous calls, with reported validation rates near 54% and congruence between algorithms less than 50%. Here, we developed an algorithm to assign a single statistic, a false discovery rate (FDR), to each somatic mutation identified by NGS. Several studies showed that Fetuin-a is an extracellular calcium-regulatory protein acting as a potent inhibitor of calcium-phosphate precipitation, involved also in bone metabolism because of its high affinity for hydroxyapatite. The aim of this study is to evaluate the serum Fetuin-A levels in patients with chronic renal failure before and after a single session of dialysis and to investigate the correlation between protein serum concentrations and T256S genotype.
