Nucleotide Variability at the Acetyl Coenzyme A Carboxylase Gene and the Signature of Herbicide Selection in the Grass Weed Alopecurus myosuroides (H…

2004

Acetyl coenzyme A carboxylase (ACCase) is the target of highly effective herbicides. We investigated the nucleotide variability of the ACCase gene in a sample of 18 black-grass (Alopecurus myosuroides [Huds.]) populations to search for the signature of herbicide selection. Sequencing 3,396 bp encompassing ACCase herbicide-binding domain in 86 individuals revealed 92 polymorphisms, which formed 72 haplotypes. The ratio of nonsynonymous versus synonymous substitutions was very low, in agreement with ACCase being a vital metabolic enzyme. Within black grass, most nonsynonymous substitutions were related to resistance to ACCase-inhibiting herbicides. Differentiation between populations was stro…

Short-term benefits, but transgenerational costs of maternal loss in an insect with facultative maternal care

2015

A lack of parental care is generally assumed to entail substantial fitness costs for offspring that ultimately select for the maintenance of family life across generations. However, it is unknown whether these costs arise when parental care is facultative, thus questioning their fundamental importance in the early evolution of family life. Here, we investigated the short-term, long-term and transgenerational effects of maternal loss in the European earwig Forficula auricularia , an insect with facultative post-hatching maternal care. We showed that maternal loss did not influence the developmental time and survival rate of juveniles, but surprisingly yielded adults of larger body and force…

Non-conventional osmotic solutes (honey and glycerol) improve mass transfer and extend shelf life of hot-air dried red carrots: Kinetics, quality, bi…

2020

The effect of solutes (sucrose, honey and glycerol) on mass transfer kinetics, physico-chemical and biological quality, and shelf life of red carrot slices (RCSs) obtained by hot-air drying at 60 degrees C/6-8 h was studied. RCSs were osmosed for 24 h in sucrose (S), sucrose + honey (SH), honey (H) and sucrose + glycerol (SGL) before drying. Honey and glycerol enhanced the water loss (WL) and solute gain (SG), giving higher values for Peleg's model constants (k(1), WL = 6.9 and 6.5, for H and SGL, respectively; k(1), SG = 92.8 and 73.5, for H and SGL, respectively). Decreased viscosity of osmotic solutions corresponded with the solute effect on WL and SG, reaching equilibrium after 16 h. Di…

A review of transgenerational effects of ocean acidification on marine bivalves and their implications for sclerochronology

2020

Abstract Ocean acidification can negatively impact marine bivalves, especially their shell mineralization processes. Consequently, whether marine bivalves can rapidly acclimate and eventually adapt in an acidifying ocean is now increasingly receiving considerable attention. Projecting the fate of this vulnerable taxonomic group is also pivotal for the science of sclerochronology – the study which seeks to deduce records of past environmental changes and organismal life-history traits from various geochemical properties of periodically layered hard tissues (bivalve shells, corals, fish otoliths, etc.). In this review, we provide a concise overview of the long-term and transgenerational respo…

Molecular Bases for Sensitivity to Acetyl-Coenzyme A Carboxylase Inhibitors in Black-Grass

2005

Abstract In grasses, residues homologous to residues Ile-1,781 and Ile-2,041 in the carboxyl-transferase (CT) domain of the chloroplastic acetyl-coenzyme A (CoA) carboxylase (ACCase) from the grass weed black-grass (Alopecurus myosuroides [Huds.]) are critical determinants for sensitivity to two classes of ACCase inhibitors, aryloxyphenoxypropionates (APPs) and cyclohexanediones. Using natural mutants of black-grass, we demonstrated through a molecular, biological, and biochemical approach that residues Trp-2,027, Asp-2,078, and Gly-2,096 are also involved in sensitivity to ACCase inhibitors. In addition, residues Trp-2,027 and Asp-2,078 are very likely involved in CT activity. Using three-…

An isoleucine residue within the carboxyl-transferase domain of multidomain acetyl-coenzyme A carboxylase is a major determinant of sensitivity to ar…

2003

Abstract A 3,300-bp DNA fragment encoding the carboxyl-transferase domain of the multidomain, chloroplastic acetyl-coenzyme A carboxylase (ACCase) was sequenced in aryloxyphenoxypropionate (APP)-resistant and -sensitive Alopecurus myosuroides (Huds.). No resistant plant contained an Ile-1,781-Leu substitution, previously shown to confer resistance to APPs and cyclohexanediones (CHDs). Instead, an Ile-2,041-Asn substitution was found in resistant plants. Phylogenetic analysis of the sequences revealed that Asn-2,041 ACCase alleles derived from several distinct origins. Allele-specific polymerase chain reaction associated the presence of Asn-2,041 with seedling resistance to APPs but not to C…

Seasonal dynamics of photosynthesis and total carbon gain in bearing and nonbearing pistachio (Pistacia vera L.) shoots

2018

Seasonal changes in leaf gas exchange, assimilation response to light and leaf area were monitored in bearing and nonbearing pistachio shoots. Shoot bearing status did not directly affect leaf photosynthetic rate. However, photosynthetic light-response curves strongly varied during the season demonstrating the dominant effect of the tree’s seasonal phenology on assimilation. Early in the season low photosynthetic rates were associated with high rates of dark respiration indicating limited photosynthesis in the young leaves. As leaves matured, dark respiration decreased and assimilation reached maximum values. Photosynthetic efficiency was strongly reduced late in the season due to leaf ag…

Universal primers for PCR-sequencing of grass chloroplastic acetyl-CoA carboxylase domains involved in resistance to herbicides

2005

Summary Primers were designed to amplify two regions involved in sensitivity to herbicides inhibiting the plastidic acetyl-CoA carboxylase (ACCase) from grasses (Poaceae). The first primer pair amplified a 551-bp amplicon containing a variable Ile/Leu codon at position 1781 in Alopecurus myosuroides sequence. The second primer pair amplified a 406-bp amplicon containing four variable codons (Trp/Cys, Ile/Asn, Asp/Gly, Gly/Ala) at positions 2027, 2041, 2078 and 2096, respectively, in A. myosuroides sequence. Both primer pairs amplified the targeted fragments from genes encoding plastidic ACCases, but not from the very similar genes encoding cytosolic ACCases. Clear DNA sequences were obtaine…

Interspecific variation of inflorescence scents and insect visitors in Allium (Amaryllidaceae: Allioideae)

2019

Allium is a large monocotyledonous genus, with many species of high economic importance. Knowledge of the pollination biology and the chemical ecology of pollination in this genus is far from being complete. We studied flower visitors of some Allium species in their native habitat and how these interaction partners communicate by olfactory cues. Floral volatiles of five Mediterranean species were investigated by dynamic headspace and thermal desorption-gas chromatography/mass spectrometry (TD-GC/MS). Floral visitors were observed and captured. The physiological activity of scent components in antennae of flower visitors and congeneric species was tested by gas chromatographic/electroantenno…

Town population size and structuring into villages and households drive infectious disease risks in pre-healthcare Finland

2021

Social life is often considered to cost in terms of increased parasite or pathogen risk. However, evidence for this in the wild remains equivocal, possibly because populations and social groups are often structured, which affects the local transmission and extinction of diseases. We test how the structuring of towns into villages and households influenced the risk of dying from three easily diagnosable infectious diseases—smallpox, pertussis and measles—using a novel dataset covering almost all of Finland in the pre-healthcare era (1800–1850). Consistent with previous results, the risk of dying from all three diseases increased with the local population size. However, the division of towns …
