A novel target of lithium therapy.
2000
Phosphatases converting 3'-phosphoadenosine 5'-phosphate (PAP) into adenosine 5'-phosphate are of fundamental importance in living cells as the accumulation of PAP is toxic to several cellular systems. These enzymes are lithium-sensitive and we have characterized a human PAP phosphatase as a potential target of lithium therapy. A cDNA encoding a human enzyme was identified by data base screening, expressed in Escherichia coli and the 33 kDa protein purified to homogeneity. The enzyme exhibits high affinity for PAP (K(m)1 microM) and is sensitive to subtherapeutic concentrations of lithium (IC(50)=0.3 mM). The human enzyme also hydrolyzes inositol-1, 4-bisphosphate with high affinity (K(m)=0…
Molecular characterization of a male-specific glycosyl hydrolase, Lma-p72, secreted on to the abdominal surface of the Madeira cockroach Leucophaea m…
2003
0264-6021 (Print) Journal Article Research Support, Non-U.S. Gov't; The epicuticular surface protein Lma-p72 is specific to the abdominal secretions of Leucophaea maderae (Madeira cockroach) adult males. Natural Lma-p72 was purified and the complete cDNA sequence determined by reverse-transcription PCR using primers based on Edman degradation fragments. Northern blot and in situ hybridization analyses showed that Lma-p72 was expressed in the tergal and sternal glands. Sequence alignment indicates that Lma-p72 is closely related to the family 1 glycosyl hydrolases (EC 3.2.1). Native Lma-p72 was proved to be active in the abdominal secretions and exhibit a beta-galactosidase-like activity. Ho…
A novel putative insect chitinase with multiple catalytic domains: hormonal regulation during metamorphosis
2002
0264-6021 (Print) Journal Article Research Support, Non-U.S. Gov't; We have used differential display to identify genes that are regulated by juvenile hormone in the epidermis of the beetle Tenebrio molitor. One of the genes encodes T. molitor chitinase 5 (TmChit5), a chitinase possessing an unusual structure. Sequence analysis of TmChit5 identified five 'chitinase units' of approx. 480 amino acids with similarity to chitinase family 18. These units are separated by less conserved regions containing putative PEST (rich in proline, glutamic acid, serine and threonine) sequences, putative chitin-binding domains and mucin domains. Northern-blot analysis identified a single transcript of approx…
Matematica ... a Distanza.
On the origin of Metazoan adhesion receptors: cloning of integrin alpha subunit from the sponge Geodia cydonium
1997
Integrins are prominent receptors known from vertebrates and the higher phyla of invertebrates. Until now, no evidence has been provided for the existence of integrins in the lowest Metazoa, the sponges (Porifera). We have isolated and characterized a cDNA clone encoding the alpha subunit of integrin from the marine sponge Geodia cydonium (GCINTEG). The open reading frame encodes a polypeptide of 1,086 residues (118 kDa). The intracellular domain features the sequence Tyr-Phe-x-Gly-Phe-Phe-x-Arg, which is different in one residue from the characteristic consensus pattern for integrin alpha subunits. We conclude that sponges, the oldest multicellular animal phylum, already utilize the struct…
The Genesis of the Italian School of Algebraic Geometry Through the Correspondence Between Luigi Cremona and Some of His Students
2023
Luigi Cremona is considered the founder of the Italian school of algebraic geometry. He formed a group of students of great value, very active in scientific research. Examining the letters from Eugenio Bertini, Ettore Caporali, and Riccardo De Paolis to Cremona preserved in the archive of the Istituto Mazziniano in Genoa, we have reconstructed their biographies, careers, studies, and relationships with their teacher. They had the merit of cultivating the scientific innovations of the period and passing them on to the subsequent generations.
An Iterative Approach to Dynamic Elastic-Plastic Analysis
1998
The step-by-step analysis of structures constituted by elastic-plastic finite elements, subjected to an assigned loading history, is here considered. The structure may possess dynamic and/or not dynamic degrees-of-freedom. As it is well-known, at each step of analysis the solution of a linear complementarity problem is required. An iterative method devoted to solving the relevant linear complementarity problem is presented. It is based on the recursive solution of a linear complementarity, problem in which the constraint matrix is block-diagonal and deduced from the matrix of the original linear complementarity problem. The convergence of the procedure is also proved. Some particular cases …
A Comparison and Survey of Finite Difference Methods for Pricing American Options Under Finite Activity Jump-Diffusion Models
2012
Partial-integro differential formulations are often used for pricing American options under jump-diffusion models. A survey on such formulations and numerical methods for them is presented. A detailed description of six efficient methods based on a linear complementarity formulation and finite difference discretizations is given. Numerical experiments compare the performance of these methods for pricing American put options under finite activity jump models.
A method for rapid generation of competitive standard molecules for RT-PCR avoiding the problem of competitor/probe cross-reactions.
1995
The analysis of gene expression is a widespread issue in a growing number of fields such as molecular genetics, immunology, and medical diagnostics. The ideal method for mRNA detection should be fast, inexpensive, sensitive, and reliable. Well-elaborated standard methods such as Northern hybridization, Sl-mapping, and RNAse protection are useful and recommended, but only reverse transcription PCR (RT-PCR) gives the highest possible sensitivity required. For many issues it is necessary not only to detect a distinct mRNA but to compare changes in mRNA levels. The use of RT-PCR for such semiquantitative and quantitative approaches resolves problems attributable to the intrinsic property of PCR…
