Cellular UDP-Glucose Deficiency Caused by a Single Point Mutation in the UDP-Glucose Pyrophosphorylase Gene

We previously isolated a mutant cell that is the only mammalian cell reported to have a persistently low level of UDP-glucose. In this work we obtained a spontaneous revertant whose UDP-glucose level lies between those found in the wild type and the mutant cell. The activity of UDP-glucose pyrophosphorylase (UDPG:PP), the enzyme that catalyzes the formation of UDP-glucose, was in the mutant 4% and in the revertant 56% of the activity found in the wild type cell. High yielding one-pot enzyme-catalyzed synthesis of UDP-glucose in gram scales

Abstract Uridine diphosphoglucose is an important cofactor of glucosylating enzymes. A simple and high yielding one-pot enzymatic synthesis of UDPG on a gram scale from glucose via hexokinase, phosphoglucomutase and UDPG pyrophosphorylase (UGPase) is described. Repetitive addition of substrate was used to avoid inhibition of UGPase. The approach allows recovery of active enzymes and their re-use. The synthesis of UDP-[4-13C]-glucose on a 0.5 g scale resulted in a final yield of 70% and a purity of >95% after chromatographic purification.

Abstract We have previously isolated a fibroblast mutant cell with high resistance to the two Rho-modifying glucosyltransferase toxins A and B of Clostridium difficile. We demonstrate here a low level of UDP-glucose in the mutant, which explains its toxin resistance since: (i) to obtain a detectable toxin B-mediated Rho modification in lysates of mutant cells, addition of UDP-glucose was required, and it promoted the Rho modification dose-dependently; (ii) high pressure liquid chromatography analysis of nucleotide extracts of cells indicated that the level of UDP-glucose in the mutant (0.8 nmol/106 cells) was lower than in the wild type (3.7 nmol/106 cells); and (iii) sensitivity to toxin B…

Measurements are reported from samples collected at twenty three sites in the Tokaj mountains of north-eastern Hungary. The simplest interpretation of the results is in terms of geomagnetic field reversals, for there is no petrological or magnetic evidence of self reversal or of correlation of oxidation state and magnetic sign. Four magnetic zones satisfy the minimum stratigraphic requirements, speculative use of these zones against theHeirtzler et al. magnetic time scale as a control suggests locating the Miocene-Pliocene boundary at 11.75 my. Warm white LED light by frequency down-conversion of mixed yellow and red Lumogen®

This work reports on the benefits and promising opportunities offered by white LED hybrid technology, based on a mixing perylene-based dyes in order to obtain a warm white light for frequency-down conversion. In a standard Ce:YAG-based white LED, the white light appears cold due to the weakness of red wavelength components in the emission spectrum. In order to obtain a warmer white, one possible solution is to add a red phosphor to the yellow one to move the chromatic coordinates properly, though the luminous efficiency drastically decreases due to the increased light absorption of the coating layer. Interannual variability of rainfall in the eastern horn of Africa and indicators of atmospheric circulation

Relationships between rainfall variations in the Eastern Horn of Africa, the Southern Oscillation, and the Indian Ocean temperature and pressure surface fields are studied for the period 1932–83. Rainfall data consist of stations and regional indices calculated for three selected areas experiencing quite different rainfall patterns. The results indicate significant negative correlations between northern autumn rains in Somalia and the Southern Oscillation during the same season. These rains are also negatively correlated with pressure in the Western Indian Ocean, and positively correlated in the Eastern Indian Ocean. The reverse pattern is shown with sea-surface temperature. Sulfur Dioxide Oxidation Catalyzed by Photosensitized Ytterbium Diphthalocyanine

Oxidation of SO2 into SO3 was found effectively catalyzed by photosensitized ytterbium diphthalocyanine, YbPc2 (Pc = phthalocyanine ligand, C32H16N8) when performed in air-saturated dimethylformamide (DMF) solution at 20 °C. The process follows according to a multi-step complex mechanism involving chemical induction as its driving force. Excitation energy of the sandwich molecular system in YbPc2 is used to promote the reaction by creating a reactive intermediate form of the ytterbium complex hosting up to eight SO2 molecules. The conversion of SO2 proved complete.
