Evidence for the existence of FGFR1-5-HT1A heteroreceptor complexes in the midbrain raphe 5-HT system.

The ascending midbrain 5-HT neurons known to contain 5-HT1A autoreceptors may be dysregulated in depression due to a reduced trophic support. With in situ proximity ligation assay (PLA) and supported by co-location of the FGFR1 and 5-HT1A immunoreactivities in midbrain raphe 5-HT cells, evidence for the existence of FGFR1–5-HT1A heteroreceptor complexes were obtained in the dorsal and median raphe nuclei of the Sprague–Dawley rat. Their existence in the rat medullary raphe RN33B cell cultures was also established. The abrogation of the HOXB7/PBX2 complex induces apoptosis in melanoma through the miR-221&222-c-FOS pathway.

Cutaneous melanoma is the fastest increasing cancer worldwide. Although several molecular abnormalities have been associated with melanoma progression, the underlying mechanisms are still largely unknown and few targeted therapies are under evaluation. Here we show that the HOXB7/PBX2 dimer acts as a positive transcriptional regulator of the oncogenic microRNA-221 and -222. In addition, demonstrating c-FOS as a direct target of miR-221&222, we identify a HOXB7/PBX2→miR-221&222 →c-FOS regulatory link, whereby the abrogation of functional HOXB7/PBX2 dimers leads to reduced miR-221&222 transcription and elevated c-FOS expression with consequent cell death. A Novel Synthetic Route to Open-Chained and Cyclic Oligoetherketones Oligoetherketones of the structure 2a,b can be generated by repetitive insertion reactions of terminal biscarbenoids 4a,b, into the OH bonds of diols like 5. The formation of the corresponding ring systems 7a and the involved carbene dimerization leading to 8b are processes which compete with the linear polyinsertion. A related one-component reaction can be performed with ω-hydroxy-1-diazo-2-alkanones (13a–c 14a–c).

Inhibitor-Induced Dimerization of an Essential Oxidoreductase from African Trypanosomes

Trypanosomal and leishmanial infections claim tens of thousands of lives each year. The metabolism of these unicellular eukaryotic parasites differs from the human host and their enzymes thus constitute promising drug targets. Tryparedoxin (Tpx) from Trypanosoma brucei is the essential oxidoreductase in the parasite's hydroperoxide-clearance cascade. In vitro and in vivo functional assays show that a small, selective inhibitor efficiently inhibits Tpx. Solvatochromic behaviour of new donor–acceptor oligothiophenes

Oligothiophene derivatives play a central role in the formulation of materials used in devices in the field of organic electronics. In this work, we report the results of the study of UV-vis absorption and fluorescence spectra, in several solvents, of a series of oligothiophenes recently synthesized in our laboratory.
