A human CCT5 gene mutation causing distal neuropathy impairs hexadecamer assembly in an archaeal model
2014
Chaperonins mediate protein folding in a cavity formed by multisubunit rings. The human CCT has eight non-identical subunits and the His147Arg mutation in one subunit, CCT5, causes neuropathy. Knowledge is scarce on the impact of this and other mutations upon the chaperone's structure and functions. To make progress, experimental models must be developed. We used an archaeal mutant homolog and demonstrated that the His147Arg mutant has impaired oligomeric assembly, ATPase activity, and defective protein homeostasis functions. These results establish for the first time that a human chaperonin gene defect can be reproduced and studied at the molecular level with an archaeal homolog. The major…
Gold nanostars coated with neutral and charged polyethylene glycols: A comparative study of in-vitro biocompatibility and of their interaction with S…
2015
Gold nanostars (GNS) have been coated with four different polyethylene glycols (PEGs) equipped with a -SH function for grafting on the gold surface. These PEGs have different chain lengths with average MW = 2000, 3000, 5000 and average number of -O-CH2-CH2 - units 44, 66, and 111, respectively. Two are neutral and two are terminated with -COOH and -NH2 functions, thus bearing negative and positive charges at physiological pH, thanks to the formation of carboxylate and ammonium groups. The negative charge of the GNS coated with PEG carboxylate has also been exploited to further coat the GNS with the PAH (polyallylamine hydrochloride) cationic polymer. Vitality tests have been carried out on …
Klīniskā farmaceita loma vispārēja tipa aptiekā
2020
The Interaction of Small Molecules with Biomolecules
2014
The binding of small molecules with biological targets is associated to interesting chemical and biological properties of the resulting supramolecular systems. We have recently reported on the synthesis and characterization of cationic first row transition metal complexes and the study of their DNA binding properties, in aqueous solutions at neutral pH, essentially investigated by viscosimetry and spectroscopic techniques such as circular dichroism, absorption and fluorescence in the UV-visible wavelength range. Of course, such procedure cannot furnish atomic level details of the molecule-DNA interaction. Computational Chemistry may provide support for the interpretation of experimental dat…
The "didactic farm" as a demonstration of school tourism from a relational point of view: an aim of business definition
2010
The article suggests different items to define the tourist service of didactic farm; moreover it compares the didactic farm with other similar services, such as: agriturism, rural tourism, ethnographic museum, etc.
Ethanol stimulates corticotropic releasing hormone (CRH) release from rat hypothalamic explants; role of acetaldehyde
2009
Ethanol is able to activate the hypothalamic-pituitary-adrenal (HPA) axis, and to modify its response to other stressors, releasing adrenocorticotropic hormone (ACTH), and glucocorticoid release, (Lee et al 1999, 2000). In the brain ethanol is diverted by catalase activity into acetaldehyde, which is reported to mediate some of its behavioural and neurochemical effects. (Peana et al 2008, Melis et al 2007) Thus, to clarifìy the mechanisms underlying ethanol activity on the HPA axis we investigated :1) ethanol effect on CRH release from incubated hypothalamic explants; 2) the role of acetaldehyde (ACD), in mediating ethanol activity. To this aim, rat hypothalamic explants were incubated with…
The effects of carnitine on the growth of sea bass, Dicentrarchus labrax L., fry
1986
Carnitine treatment has an appreciable effect on the hatchery-reared sea bass, Dicentrarchus labrax, fry. The administration of the laevorotatory isomer (1-carnitine) which is engaged in the transport of the acyl and acetyl groups through the mitochondrial membrane, stimulating lipid metabolism, increases the growth rate and the protein content of the treated fry. The dextrorotatory isomer (d-carnitine), which is an antagonist of the 1-carnitine, has an opposite effect on the growth and metabolism of the treated fry.
Long-term effects of contrasting tillage on soil organic carbon, nitrous oxide and ammonia emissions in a Mediterranean Vertisol under different crop…
2018
This 2-year study aimed to verify whether the continuous application of no tillage (NT) for over 20 years, in comparison with conventional tillage (CT), affects nitrous oxide (NO) and ammonia (NH) emissions from a Vertisol and, if so, whether such an effect varies with crop sequence (continuous wheat, WW and wheat after faba bean, FW). To shed light on the mechanisms involved in determining N-gas emissions, soil bulk density, water filled pore space (WFPS), some carbon (C) and nitrogen (N) pools, denitrifying enzyme activity (DEA), and nitrous oxide reductase gene abundance (nosZ gene) were also assessed at 0–15 and 15–30 cm soil depth. Tillage system had no significant effect on total NH e…
Alcohol preference, behavioural reactivity and cognitive functioning in female rats exposed to a three-bottle choice paradigm.
2012
Alcohol abuse is a substantial and growing health problem in Western societies. In the last years in vivo and in vitro studies have suggested that males and females display a different alcohol drinking behaviour, with swingeing differences not only in the propensity for alcohol use but also in the metabolic and behavioural consequences. In this study we investigated, in adult female rats, ethanol self-administration and preference pattern using a 3-bottle paradigm with water, 10% ethanol solution, and white wine (10%, v/v), along a four-week period. The influence of alcohol free-access on explorative behaviour in the open field (OF), and on spatial learning and reference memory in the Morri…
