Abstract LB-017: HSP110 sustains aberrant NFkB signaling in activated B-cell diffuse large B-cell lymphoma through MyD88 stabilization

2017

Abstract Diffuse large B cell lymphoma (DLBCL) is an aggressive lymphoproliferative disorder of B lymphocytes accounting for 30 % of adult Non Hodgkin Lymphoma (NHL). Among DLBCL, Activated B Cell - DLBCL (ABC-DLBCL) is the most aggressive form and has a poor prognosis. Heat-shock proteins (HSPs) are molecular chaperons highly expressed in cancer cells and implicated in resistance to radio- and chemotherapy. Therefore, HSPs are envisioned as therapeutic targets in many cancers. Among the different HSPs, HSP110 has been recently identified as a pro-survival factor in germinal center-derived DLBCL (GC-DLBCL), through stabilization of the GC-DLBCL oncogene Bcl-6. Here, we have explored if HSP1…

On the Effect of Downscaling in Inkjet Printed Life-Inspired Compartments

2019

The fabrication of size-scalable liquid compartments is a topic of fundamental importance in synthetic biology, aiming to mimic the structures and the functions of cellular compartments. Here, inkjet printing is demonstrated as a customizable approach to fabricate aqueous compartments at different size regimes (from nanoliter to femtoliter scale) revealing the crucial role of size in governing the emerging of new properties. At first, inkjet printing is shown to produce homogenous aqueous compartments stabilized by oil-confinement with mild surfactants down to the hundreds of picoliter scale [1]. Raster Image Correlation Spectroscopy allows to monitor few intermolecular events by the involv…

Influence of Pigmentation on the Codeine Content of Hair Fibers in Guinea Pigs

1997

Tortoise shell guinea pigs (n = 7) were administered codeine (1 mg/mL codeine-base) in their drinking water for 3 weeks. Black, reddish-brown and white hair was collected separately from each animal before and after treatment. The hair samples were analyzed by GC/MS. The experiment showed positive results for all hair fibers with large individual variability of drug incorporation. Low drug intake resulted in small differences of the drug content in hair fibers different in color, whereas in cases of high drug intake a strong influence of hair pigmentation on the analytical results was observed. The highest drug content was always found in black hair samples, non-pigmented hair showed the lo…

Guava (Psidium guajava L.) Leaf Extract as Bioactive Substances for Anti-Androgen and Antioxidant Activities

2022

Leaves of guava (Psidium guajava L.) have been used in Thai folk medicine without any supporting evidence as a traditional herbal remedy for hair loss. Androgenetic alopecia (AGA) is chronic hair loss caused by effects of androgens in those with a genetic predisposition, resulting in hair follicle miniaturization. Our objectives were to provide the mechanistic assessment of guava leaf extract on gene expressions related to the androgen pathway in well-known in vitro models, hair follicle dermal papilla cells (HFDPC), and human prostate cancer cells (DU-145), and to determine its bioactive constituents and antioxidant activities. LC-MS analysis demonstrated that the main components of the et…

The impact of systemic and illiquidity risk on financing with risky collateral

2015

Abstract Repurchase agreements (repos) are one of the most important sources of funding liquidity for many financial investors and intermediaries. In a repo, some assets are given by a borrower as collateral in exchange of funding. The capital given to the borrower is the market value of the collateral, reduced by an amount termed as haircut (or margin). The haircut protects the capital lender from loss of value of the collateral contingent on the borrower׳s default. For this reason, the haircut is typically calculated with a simple Value at Risk estimation of the collateral for the purpose of preventing the risk associated to volatility. However, other risk factors should be included in th…

Magnetic coiffure: Engineering of human hair surfaces with polyelectrolyte-stabilised magnetite nanoparticles

2022

Here we report a spontaneous electrostatic coating of human hair with aqueous Fe3O4 colloids capable to tailor magnetic properties to hair, orienting and even moving them under the influence of the external magnetic field. Magnetite particles were modified by cationic and anionic polyelectrolytes and then successfully deposited in dense arrays, starting from cuticle gaps and spreading further over a major hair surface. These biocompatible and biodegradable magnetic nanoparticles may serve as carriers for drug loading and delivery for topical pharmaceutical treatments. The deposition process was imaged in real-time using dark-field microscopy. The hair specimens were further studied using a …

Mutated cylindromatosis gene affects the functional state of dendritic cells

2010

Cylindromatosis gene (CYLD) is a ubiquitously expressed deubiquitinating enzyme, which interacts with members of the NF-κB signaling pathway and attenuates NF-κB and JNK signaling. Here, we report that DC derived from transgenic mice, which solely express a naturally occurring CYLD isoform (CYLD(ex7/8)), display a higher content of nuclear RelB and express elevated levels of NF-κB family members as well as of known NF-κB-target genes comprising costimulatory molecules and pro-inflammatory cytokines, as compared with WT DC. Accordingly, unstimulated CYLD(ex7/8) DC exhibited a significantly higher primary allogenic T-cell stimulatory capacity than WT DC and exerted no tolerogenic activity. Tr…

Conserved and newly acquired roles of PIF1 homologs in tomato (Solanum lycopersicum)

2021

ABSTRACTPHYTOCHROME INTERACTING FACTORS (PIFs) are transcription factors that interact with the photoreceptors phytochromes and integrate multiple signaling pathways related to light, temperature, defense and hormone responses. PIFs have been extensively studied inArabidopsis thaliana, but less is known about their roles in other species. Here, we investigate the role of the two homologs of PIF1 found in tomato (Solanum lycopersicum), namely PIF1a and PIF1b. Analysis of gene expression showed very different patterns, indicating a potential evolutionary divergence in their roles. At the protein level, light regulated the stability of PIF1a, but not PIF1b, further supporting a functional dive…

Adhesive root hairs facilitate Posidonia oceanica seedling settlement on rocky substrates

2015

Posidonia oceanica, the dominant Mediterranean seagrass, has been historically described as a species typically growing on mobile substrates whose development requires precursor communities. During more than 10 years of direct observations, we noticed that P. oceanica seedlings were often firmly anchored to rocky reefs, even at exposed sites. Thus, we analysed the ultrastructural features of seedling root systems to identify specific traits that may represent adaptations for early seedling anchorage on rocky bottoms. Subapical sections of adventitious roots were obtained from 2-3 months old specimens collected in the field and were observed at SEM revealing an extensive coverage of adhesive…

Trace elements in hair of urban schoolboys: a diagnostic tool in environmental risk assessment.

2011

Hair analysis may represent a means to quantify the relationship between human exposure to metal contamination and the environmental conditions of workplaces or residence sites. Hair are stable and their composition does not change over short time period. Furthermore, sampling procedure is very easy, requires no specific professional skills, is painless and non-invasive. 334 hair samples were collected from Caucasian children 11-13 years old, of both genders, without colored or treated hair, living in several Sicilian towns characterized by different geochemical environments: urban (Palermo), volcanic (Etna), mining area (Antillo-Fiumedinisi), industrial site (Pace del Mela), uncontamined a…
