Molecular mechanisms linking amyloid β toxicity and Tau hyperphosphorylation in Alzheimer׳s disease

2015

Neurofibrillary tangles (aggregates of cytoskeletal Tau protein) and senile plaques (aggregates mainly formed by amyloid β peptide) are two landmark lesions in Alzheimer׳s disease. Some researchers have proposed tangles, whereas others have proposed plaques, as primary lesions. For a long time, these were thought of as independent mechanisms. However, experimental evidence suggests that both lesions are intimately related. We review here some molecular pathways linking amyloid β and Tau toxicities involving, among others, glycogen synthase kinase 3β, p38, Pin1, cyclin-dependent kinase 5, and regulator of calcineurin 1. Understanding amyloid β and Tau toxicities as part of a common pathophys…

Thioflavin T templates amyloid β(1–40) conformation and aggregation pathway

2015

Aβ(1-40) peptide supramolecular assembly and fibril formation processes are widely recognized to have direct implications in the progression of Alzheimer's disease. The molecular basis of this biological process is still unknown and there is a strong need of developing effective strategies to control the occurring events. To this purpose the exploitation of small molecules interacting with Aβ aggregation represents one of the possible routes. Moreover, the use specific labeling has represented so far one of the most common and effective methods to investigate such a process. This possibility in turn rests on the reliability of the probe/labels involved. Here we present evidences of the effe…

A Population-Based 3D Atlas of the Pathological Lumbar Spine Segment

2022

The spine is the load-bearing structure of human beings and may present several disorders, with low back pain the most frequent problem during human life. Signs of a spine disorder or disease vary depending on the location and type of the spine condition. Therefore, we aim to develop a probabilistic atlas of the lumbar spine segment using statistical shape modeling (SSM) and then explore the variability of spine geometry using principal component analysis (PCA). Using computed tomography (CT), the human spine was reconstructed for 24 patients with spine disorders and then the mean shape was deformed upon specific boundaries (e.g., by ±3 or ±1.5 standard deviation). Results dem…

Muscle Dysmorphia in Adolescence: The Role of Parental Psychological Control on a Potential Behavioral Addiction

2019

Objective The study investigated the relationship between psychological parental control and muscle dysmorphia in adolescence, as form of exercise dependence, focusing also on the role of pathological worry. Methods Participants were 312 adolescents (140 boys and 172 girls) aged 16 to 18 years (M= 17.05; SD = 0.85) and completed the Muscle Dysmorphia Disorder Inventory, the Dependency-oriented and Achievement-oriented Parental Psychological Control, and the Penn State Worry Questionnaire. Results The results highlighted that boys showed higher level than girls in dependency-oriented and achievement-oriented parental psychological control and muscle dysmorphia. Furthermore, girls showed high…

Summary ― The work presents the results of the bio-archaeological analyzes carried out on human skeletal remains found in fourteen burials of Medieval chronology at the Archaeological Park of Valle dei Templi in Agrigentum (Sicily). The aim of the work was the acquisition of the information necessary for the reconstruction of the biological profile of each individual throught the determination of sex the estimate of the biological age at death the estimate of height and the evaluation of pathologies and markers of occupational stress through current anthropological diagnostic methodologies and techniques. Althought the poor state of conservation of some individuals has not allowed their whole anthropological characterization the investigations have highlighted the heterogeneity relating to the age groups and have allowed us to ascertain the presence of pathological alterations in subjects of mature age sometimes of unknown etiology such as DISH (Diffuse Idiopathic Hyperostosis). The sample size is not representative of the entire population but subsequent insights will provide a better understanding of the population dynamics of Medieval Agrigentum.Settore BIO/08 - Antropologia

Predictors of early dropout in treatment for gambling disorder: The role of personality disorders and clinical syndromes

2017

Several treatment options for gambling disorder (GD) have been tested in recent years; however dropout levels still remain high. This study aims to evaluate whether the presence of psychiatric comorbidities predicts treatment outcome according to Millon's evolutionary theory, following a six-month therapy for GD. The role of severity, duration of the disorder, typology of gambling (mainly online or offline) and pharmacological treatment were also analysed. The recruitment included 194 pathological gamblers (PGs) to be compared with 78 healthy controls (HCs). Psychological assessment included the South Oaks Gambling Screen and the Millon Clinical Multiaxial Inventory-III. The "treatment fail…

Influence of polysaccharides on wine protein aggregation.

2016

Abstract Polysaccharides are the major high-molecular weight components of wines. In contrast, proteins occur only in small amounts in wine, but contribute to haze formation. The detailed mechanism of aggregation of these proteins, especially in combination with other wine components, remains unclear. This study demonstrates the different aggregation behavior between a buffer and a model wine system by dynamic light scattering. Arabinogalactan-protein, for example, shows an increased aggregation in the model wine system, while in the buffer system a reducing effect is observed. Thus, we could show the importance to examine the behavior of wine additives under conditions close to reality, in…

Golgi Fragmentation in Neurodegenerative Diseases: Is There a Common Cause?

2019

In most mammalian cells, the Golgi complex forms a continuous ribbon. In neurodegenerative diseases, the Golgi ribbon of a specific group of neurons is typically broken into isolated elements, a very early event which happens before clinical and other pathological symptoms become evident. It is not known whether this phenomenon is caused by mechanisms associated with cell death or if, conversely, it triggers apoptosis. When the phenomenon was studied in diseases such as Parkinson’s and Alzheimer’s or amyotrophic lateral sclerosis, it was attributed to a variety of causes, including the presence of cytoplasmatic protein aggregates, malfunctioning of intracellular traffic and/or alterations i…
