Identification of Novel Principles of Keratin Filament Network Turnover in Living Cells

It is generally assumed that turnover of the keratin filament system occurs by exchange of subunits along its entire length throughout the cytoplasm. We now present evidence that a circumscribed submembranous compartment is actually the main site for network replenishment. This conclusion is based on the following observations in living cells synthesizing fluorescent keratin polypeptides: 1) Small keratin granules originate in close proximity to the plasma membrane and move toward the cell center in a continuous motion while elongating into flexible rod-like fragments that fuse with each other and integrate into the peripheral KF network. 2) Recurrence of fluorescence after photobleaching i…

The chain dynamics in supramolecular polymer networks is determined by the interplay of the kinetics of transient interchain association and relaxation of the network chains themselves. This interplay can be addressed by studying model supramolecular polymer networks in which the number of associative side groups and the molar mass of the covalently jointed backbone polymers are both varied systematically. Publisher Summary The characterization and development of autofluorescent proteins, most prominently of the green florescent protein, have provided tools to label cellular structures such that they can be examined in living cells. This chapter highlights the potential of live cell imaging in providing novel and unprecedented insights into the dynamic organization of the keratin cytoskeleton and outlines the important aspects of this method. Preservation in Disaster Situations: a case study of the Valvilla Wool Mill Museum, Finland

This article discusses the role of documentation and rescue planning in minimizing collection damages in disaster situations. The topic is surveyed through a case study of a fire that occurred in 2003 at the Valvilla Wool Mill museum in Hyvinkää, in Southern Finland. This incident caused significant damage to the authenticity and integrity of the museum’s archival collection. Both the fire and the extinguishing of the fire caused severe damage to items in the archival collection. Hydraulic turbines for energy recovery in wastewater treatment plants, with relatively large discharges values and small head jumps, are usually screw Archimedes or Kaplan types. In the specific case of a small head jump (about 3 m) underlying a rectangular weir in the major Palermo (Italy) water treatment plant, a traditional Kaplan solution is compared with two other new proposals: a Hydrostatic Pressure Machine (HPM) located at the upstream channel and a cross-flow turbine (CFT) located in a specific underground room downstream of the same channel.
