Inter-individual variability of protein patterns in saliva of healthy adults

International audience; In order to document inter-individual variability in salivary protein patterns, unstimulated whole saliva was obtained from 12 subjects at 10 am and 3 pm of the same day. Saliva proteins were separated using two-dimensional gel electrophoresis, and semi-quantified using image analysis. One-way ANOVA was used to test the effects “time of sampling” and “subject”. Data were further explored by multivariate analyses (PCA, hierarchical clustering). Spots of interest were identified by mass spectrometry (MALDI-TOF MS/MS and nanoLC ESI-IT MS/MS). Between 1986 and 1991 approximately 6500 Latvian inhabitants were recruited for clean-up work at the Chernobyl nuclear power plant. Their absorbed doses are usually unknown, because less than half of them had their external exposure officially documented. Clinical investigations show a high morbidity rate for these clean-up workers when compared with that of the general population. In order to understand the causes of their diseases and the impact of ionising radiation, electron spin resonance (ESR) has been used to measure the absorbed doses in human tooth enamel. In the latter conditions, two different sampling points (t = 0 and t = 10 min) corresponding with oral (25.5 °C) and postoral phases (36 °C) were monitored. We developed and evaluated two different strategies for determining abuse drugs based on (i) the analysis of saliva by ion mobility spectrometry (IMS) after thermal desorption and (ii) the joint use of IMS and infrared (IR) spectroscopy after liquid-liquid microextraction (LLME) to enable the sensitivity-enhanced detection and double confirmation of ecstasy (MDMA) abuse. Both strategies proved effective for the intended purpose.
