Injectable hydrogel formulations to host adipose stem cell spheroids for stemness maintenance and bone and cartilage regeneration

2021

Adipose-derived mesenchymal stem cells (ASCs) represent a great promise for tissue regeneration as fat is a very abundant source of stem cells (1) and owing to their ability to self-renew and differentiate into specific tissue types (2). In general, they are usually cultured as two-dimensional (2D) adherent monolayers, not representative of the in vivo condition, often entailing lower cell viability and, more in general, a lower “cell quality” in terms of regenerative potential (3, 4). When ASCs are cultured in low adhesion flasks and with a suitable culture medium, they aggregate in the form of three-dimensional spheroids (SASCs). The incorporation of these spheroids into injectable, in-si…

Biological characterization of Poly-L-lactic acid (PLLA)/Hydroxyapatite (HA)/Bioglass (BG) composite scaffolds made by Thermally Induced Phase Separa…

In the last few years, Tissue Engineering has focused on the favourable effects that composite scaffolds have on cell adhesion, growth and differentiation. In fact, composite scaffolds, usually composed of a synthetic polymer supplemented with naturally occurring components, display superior mechanical properties and bioconductivity than scaffolds consisting of a single component. Hydroxyapatite (HA) is the major inorganic component of bones. Bioglass (BG) is known to exert stimulatory effects on cells by ion release and hence, could be also advantageous for Bone Tissue Engineering. Poly-L-lactic acid (PLLA) is a versatile synthetic polymer combinable with HA and BG. The aim of this work wa…

OBTAINING MESENCHYMAL STEM CELLS FROM ADIPOSE TISSUE OR MURIN ORIGIN: EXPERIMENTAL STUDY.

2014

The aim of this study was to isolate and characterize rat adipose Derived Mesenchymal Stem Cells (AD-MSCs) in order to evaluate their proliferative potential and their ability to different cell types. AD-MSCs and Derived Mesenchymal Stem Cells (BM-MSCs) have the same characteristic in terms of plasticity. The advantage of adipose tissue is that it is an easier accessible source and it offers a large amount of MSCs by less invasive surgical tecniques. MSCs were obtained from subcutaneous adipose tissue of Wistar rats. first of all microbiological controls were made to exclude the presence of bacteria of fungi in then tissue. Adipose tissue was mechanically and enzimatically fragmented and st…

MESENCHIMAL STEM CELLS IN INFLAMMATORY BOWEL DISEASES: CLINICAL EVIDENCES AND POTENTIAL INSIGHTS FOR THE CLINICIANS

2013

Mesenchimal stem cells (MSCs) have been used experimentally and clinically in the treatment of a wide variety of pathologies MSCs can be safely transplanted in autologous and allogenic ways as they are non-immunogenic, and consequently represent a therapeutic option for refractory connective tissue diseases, fibrosing diseases like scleroderma and fistulizing colitis like in Crohn's disease (CD). The immunomodulatory properties of MSCs have already shown promise when used as therapy for otherwise medically refractory CD. Accumulating evidence suggests that the properties may also be exploited of several other conditions. The currently available experimental and clinical data indicate that, …

NUTRITION IN IBD PATIENTS

2013

Inflammatory Bowel Disease (IBD) is a chronic disorder characterized by a relapsing-remitting course, which alternates between active and quiescent states, ultimately impairing a patients' quality of life. The two main types of IBD are Crohn's disease (CD) and Ulcerative Colitis (UC). CD Shows a transmural granulomatous inflammation that can involve any segment of the intestine affecting all layers of the intestinal wall, while UC is limited to the mucosa and superficial submucosa of the colon. In physiological conditions the gut is costantly exposed to various antigens, commensal microflora and pathogens and the inflammatory response is finely balanced. Anyhow i some individuals with geneti…

Obtaining mesenchymal stem cells from adipose tissue of murine origin: Experimental study

2015

Stem cells have a key role in regenerative medicine and tissue engineering. Although not immortal, they are able to expand manyfold in culture retaining at the same time their growth and multilineage potential. They also show a migratory capacity when transplanted systemically in animal model with injuries. Thanks to their properties and their plasticity stem cells are of great importance since they can be used as a tool for repair damaged tissues and organs. Mesenchymal stem cells, in particular, have the ability to differentiate into lineages of mesodermal tissues, such as skeletal muscle, bone, tendons, cartilage, and fat under appropriate culturing conditions. Recent evidence suggest th…

INTRODUCING NON-ADHERENT PROGENITORS FROM ADIPOSE-DERIVED STEM CELLS (NAPADSCS): PROOF OF STEMNESS AND POSSIBLE FUTURE APPLICATIONS IN REGENERATIVE S…

2012

Introduction: Mesenchymal Stromal Cells (MSCs) can be isolated from adipose tissue and are currently used in vitro for experimental studies. MSCs are often mislabeled as Adipose-derived Stem Cells (ADSCs) despite presenting a more differentiated phenotype. The isolation and expansion of non-adherent progenitors from adipose-derived stem cells (napADSCs), has been achieved in our laboratory and is currently under investigation. In this study we investigate: a) the proof of stemness of such progenitors, and b) the feasibility of napADSCs adhesion over Integra® for cell colonization and for future differentiation and engineering of semi-synthetic tissues. Material and Methods: Adipose tissue (…
