In human osteosarcoma 3AB-OS cancer stem cells let-7d seems to have the dual function of oncogene/tumor-suppressor miRNA

2015

Introduction: Osteosarcoma (OS), an aggressive tumor a!ecting adolescents, shows therapy resistance and recurrence which can depend on cancer stem cells (CSCs), the source for tissue renewal and hold malignant potential; thus, OS treatment requires their eradication. Here, using 3AB-OS CSCs previously obtained from the OS-MG63 cells, we focused on the role of let-7d in managing stemness properties of 3AB-OS CSCs. Methods: 3AB-OS CSCs were stably transfected with pCDomH-plasmid, containing mir-let7-d, or empty vector as a control. Cell proliferation and viability were evaluated by EdU and Trypan blue assays, respectively. Sarcosphere and colony assays and wound healing and transwell invasion…

Burst analysis tool for developing neuronal networks exhibiting highly varying action potential dynamics

2012

In this paper we propose a firing statistics based neuronal network burst detection algorithm for neuronal networks exhibiting highly variable action potential dynamics. Electrical activity of neuronal networks is generally analyzed by the occurrences of spikes and bursts both in time and space. Commonly accepted analysis tools employ burst detection algorithms based on predefined criteria. However, maturing neuronal networks, such as those originating from human embryonic stem cells (hESCs), exhibit highly variable network structure and time-varying dynamics. To explore the developing burst/spike activities of such networks, we propose a burst detection algorithm which utilizes the firing …

Development of Bioimage Analysis tools for the study of neurogenesis and neurodegeneration

2021

Rat Cardiac progenitor cells and their application in cell therapy

2015

cells and the subsequent heart failure. When the pharmacological approach no longer complies with the disease evolution, organ transplantation appears to be the only treatment able to rescue the patient life. Cell therapy promises to be clinically efficient and would allow circumventing many limitations of organ transplantation, such as organ low availability, major surgical procedures, high costs and longterm immunosuppression [1]. We designed porous Poly-Lactic Acid (PLLA) and Fibroin scaffolds to deliver CPCs in the heart, we isolated and characterized CPCs for the expression of c-Kit, MDR-1 and Sca-1 by flow cytometry, we tested their degree of differentiation in vitro studying the expr…

Skin Immunomodulation during Regeneration: Emerging New Targets

2021

Adipose-Derived Stem Cells (ADSC) are present within the hypodermis and are also expected to play a pivotal role in wound healing, immunomodulation, and rejuvenation activities. They orchestrate, through their exosome, the mechanisms associated to cell differentiation, proliferation, and cell migration by upregulating genes implicated in different functions including skin barrier, immunomodulation, cell proliferation, and epidermal regeneration. ADSCs directly interact with their microenvironment and specifically the immune cells, including macrophages and T and B cells, resulting in differential inflammatory and anti-inflammatory mechanisms impacting, in return, ADSCs microenvironment and …

Hopes and Limits of Adipose-Derived Stem Cells (ADSCs) and Mesenchymal Stem Cells (MSCs) in Wound Healing

2020

Adipose tissue derived stem cells (ADSCs) are mesenchymal stem cells identified within subcutaneous tissue at the base of the hair follicle (dermal papilla cells), in the dermal sheets (dermal sheet cells), in interfollicular dermis, and in the hypodermis tissue. These cells are expected to play a major role in regulating skin regeneration and aging-associated morphologic disgraces and structural deficits. ADSCs are known to proliferate and differentiate into skin cells to repair damaged or dead cells, but also act by an autocrine and paracrine pathway to activate cell regeneration and the healing process. During wound healing, ADSCs have a great ability in migration to be recruited rapidly…
