Synthesis and Biological Properties of Benzothiazole, Benzoxazole, and Chromen-4-one Analogues of the Potent Antitumor Agent 2-(3,4-Dimethoxyphenyl)-…

New fluorinated 2-aryl-benzothiazoles, -benzoxazoles, and -chromen-4-ones have been synthesized and their activity against MCF-7 and MDA 468 breast cancer cell lines compared with the potent antitumor benzothiazole 5. Analogues such as 9a, b and 12a, d yielded submicromolar GI50 values in both cell lines; however, none of the new compounds approached 5 in terms of antitumor potency. For 5, binding to the aryl hydrocarbon receptor appeared to be necessary but not sufficient for growth inhibition.

Between the idea and the construction of the architecture the drawing plays a central role to connect the different protagonists of a project. It involves the community by envisaging unprecedented perspectives. Writing is useful to explicit the graphics and establish a new order in the design reasoning. The relationships between signs and meanings multiply in recent information systems in which it seems possible to conclude a lot (thanks to numerical and spatial data) but sometimes a synthesis is missing though that synthesis is fundamental to create new physical experiences. For the exploration of the use of different architectural languages and for the push towards the project rather than a sterile cataloging the case of the Piano Programma for the historic center of Palermo (Giuseppe Samonà Giancarlo De Carlo Umberto Di Cristina Anna Maria Sciarra Borzí 1979-82) reveals to be ambitious. Giuseppe Samonà integrates the classical representation based on Monge's projections with photographs and iconic drawings associating signs with a linguistic-structural communication capable of expressing the relationships between the parts at the basis of a morphological approach to the plan. The graphic story ("second language") of the urban fabric has creative implications expressed by images and annotations ("first language") useful for decoding them. Since the plan focuses on the potential of a storytelling that orients the imagination of its readers (designers citizens politicians) it can contribute to the study titled "B4R– BRANDING 4 RESILIENCE - Tourist infrastructure as a tool to enhance small villages by drawing resilient communities and new open habitats” as a starting point for the mapping in which GIS analysis and multidisciplinary data (geometric topological and quantitative) relating to some villages of the internal Sicily shall translate into a new architectural urban and landscape quality.Settore ICAR/14 - Composizione Architettonica E Urbana

Synthesis and biological evaluation of new bicyclic fluorinated uracils through ring-closing metathesis.

Two families of bicyclic fluorinated uracils have been prepared starting from a gem-difluorinated unsaturated nitrile, by means of a ring-closing metathesis reaction to form the new ring, which is fused at the C-5/C-6 or N-1/C-6 positions of the uracil moiety. The selective formation of olefin regioisomers in the metathesis process can be controlled according to the reaction conditions (catalyst, solvent, and temperature). The acaricidal activities of the resulting compounds have also been investigated.

Auxiliary-controlled stereoselective enolate protonation: Enantioselective synthesis of cis and trans annulated decahydroquinoline alkaloids

Abstract The diastereoselective synthesis of the octahydroquinoline enone precursor of pumiliotoxin C is achieved via tandem Mannich-Michael reaction on N-galactosyl imines. Conjugate cuprate addition to the bicyclic enone stereoselectively forms the trans annulated 4a- epi -pumiliotoxin C skeleton in the presence of the carbohydrate auxiliary, and the cis annulated pumiliotoxin C skeleton in its absence.

Preparation of dopaminergic N-alkyl-benzyltetrahydroisoquinolines using a 'one-pot' procedure in acid medium.

The preparation of N-methyl-BTHIQ (4) from N-phenylethyl-phenacetamide (1) by cyclization, reduction and N-alkylation in acid medium has been achieved in good yield in a 'one-pot' procedure. Acylation of imine (2) intermediate afforded the Z and E stereoselectivity in the enamide formation. 6-Hydroxy-BTHIQ (7) shows selectivity for D2 dopamine receptors, while its N-methylated homologue (8) displays higher affinities for both D1 and D2 receptor types, with an unexpected increase in D1 dopamine receptor affinity.

In an effort to learn more about the general structure-activity relationships of cocaine with the aim to elucidate those structural features that might confer antagonistic properties to such analogues, we describe herein our synthetic efforts to prepare two-carbon bridge functionalized (methoxylated and hydroxylated) analogues. Our approach makes use of a modification of the classical Willstatter synthesis of cocaine: Mannich type cyclization of acetonedicarboxylic acid monomethyl ester with methylamine hydrochloride and 2-methoxysuccindialdehyde in a citrate buffer solution afforded the 6- and 7-substituted 2-carbomethoxy-3-tropinones 3a,b and 4a,b in approximate yields of 64%. In the series described in this work, the hydrothermal synthesis led to oxidation of the 5-methyl-pyrazinecarboxylate anion to the 2,5-pyrazinedicarboxylate dianion (2,5-pzdc) allowing the preparation of three-dimensional (3D) lanthanide(III) organic frameworks of formula {[Ln2(2,5-pzdc)3(H2O)4]·6H2O}n [Ln = Ce (1), Pr (2), Nd (3), and Eu (4)] and {[Er2(2,5-pzdc)3(H2O)4]·5H2O}n (5). Single-crystal X-ray diffraction on 1–5 reveals that they crystallize in the triclinic system, P1 space group with the series 1–4 being isostructural. Abstract The enantioselective epoxide biohydrolysis of the racemic benzodioxole 6,7-epoxygeraniol derivative 1 has been achieved using the fungus A. niger . This new type of preparative scale bioconversion allows the synthesis of both enantiomers of Bower's compound, an analogue of insect juvenile hormone. Biological tests showed that the 6(R) enantiomer was about ten times more active than the 6(S) enantiomer against the yellow meal worm Tenebrio molitor .
