3.13 Carbohydrate Derived Auxiliaries: Amino Sugar and Glycosylamine Auxiliaries

2012

The stereodifferentiating potential of carbohydrate auxiliaries is illustrated by examples of oxazolidinones derived from amino sugars and aldimines of glycosylamines. Taking advantage of the bicyclic structure of amino sugar-derived oxazolidinones, reactions of their unsaturated or deprotonated N -acyl derivatives show an efficient exo - versus endo -differentiation. High diastereoselectivity is achieved in the syntheses of β -branched or α -substituted carboxylic acid derivatives. Imines of glycosylamines, in particular of 2,3,4,6-tetra- O -pivaloyl- β - d -galactopyranosylamine, are characterized by a conformation stabilized through the exo -anomeric effect and by their ability to coordi…

Synthetic MUC1 Antitumor Vaccine Candidates with Varied Glycosylation Pattern Bearing R/S-configured Pam3 CysSerLys4.

2016

The Toll-like receptor 2 ligand Pam3 CysSer is of particular interest for the construction synthetic vaccines because of its ability to stimulate of the innate immune system. Such vaccines usually comprise Pam3 CysSer with the natural R-configuration at the glycerol 2-position. Pam3 CysSer peptide vaccines with natural configuration have been shown to be more efficient than the corresponding R/S diastereomers. In order to clarify whether the effect of the configuration of Pam3 Cys on the immune response also applies to glycopeptide vaccines, MUC1 glycopeptide-lipopeptide vaccines bearing either R- or R/S-configured Pam3 CysSerLys4 were compared for their immunological effects. In order to f…

Synthesis of Tn/T Antigen MUC1 Glycopeptide BSA Conjugates and Their Evaluation as Vaccines

2011

The tumor-associated mucin MUC1 over-expressed in most epithelial tumor tissues is considered a promising target for immunotherapy. The extracellular part of MUC1 contains a domain of numerous tandem repeats of the amino acid sequence HGVTSAPDTRPAPGSTAPPA, including five potential O-glycosylation sites. In this study, T9 and S15 have been chosen as the positions of glycosylation. The glycopeptides N-terminally equipped with a triethylene glycol spacer were synthesized by microwave-assisted Fmoc solid-phase peptide synthesis. After detachment from the resin and deprotection, the MUC1 glycopeptides were conjugated to bovine serum albumin (BSA). To evaluate the immunological properties, balb/c…

Herbicidal derivatives of aminomethylenebisphosphonic acid. Part III. Structure-activity relationship.

1997

Derivatives of aminomethylenebisphosphonic acids constitute a class of promising herbicides. More than 40 N-substituted aminomethylenephosphonic acids were synthesized and evaluated for their herbicidal activity on common cress (Lepidium sativum L.) and cucumber (Cucumis sativus L.). Some of the tested compounds were found to exhibit strong herbicidal properties being equal in activity with the popular herbicide glyphosate as well as parent N-pyridylaminomethylenephosphonic acids. N-Substituted iminodi(methylenephosphonic) acids, which may be considered as close analog of glyphosate, were inactive toward test plants.

Functionnalized magnetite nanoparticles for bimodal imaging MRI/PET

2015

Functionalized magnetite nanoparticles for bimodal MRI/PET imagingSuperParamagnetic Iron Oxide Nanoparticles (SPIONs) have been widely studied in the biomedical field due to their promising application as nanodrugs and MRI (Magnetic Resonance Imaging) contrast agents (T2). In this study, magnetite (Fe3O4) nanoparticles have been developed for use as contrast agents for MRI/PET (Positron emission tomography) double imaging. First, functionalized stable superparamagnetic SPIONs have been synthesized in a continuous hydrothermal reactor. During the synthesis, hydrophilic agents (citric acid, LDOPA, DHCA and PHA) have been grafted on the surface of the nanoparticles. The functionalization of th…

Xylochemistry--Making Natural Products Entirely from Wood.

2015

The first total synthesis of the dimeric berberine alkaloid ilicifoline (ilicifoline B) is reported. Its carbon skeleton is constructed from ferulic acid, veratrole, and methanol. The synthesis reported herein employs starting materials solely derived from wood. The natural product is thus constructed entirely from renewable resources. The same strategy is applied to a formal total synthesis of morphinan alkaloids. The use of wood-derived building blocks (xylochemicals) instead of the conventional petrochemicals represents a sustainable alternative to classical synthetic approaches.

Iron-Catalyzed C−O Bond Activation: Opportunity for Sustainable Catalysis

2017

Oxygen-based electrophiles have emerged as some of the most valuable cross-coupling partners in organic synthesis due to several major strategic and environmental benefits, such as abundance and potential to avoid toxic halide waste. In this context, iron-catalyzed C-O activation/cross-coupling holds particular promise to achieve sustainable catalytic protocols due to its natural abundance, inherent low toxicity, and excellent economic and ecological profile. Recently, tremendous progress has been achieved in the development of new methods for functional-group-tolerant iron-catalyzed cross-coupling reactions by selective C-O cleavage. These methods establish highly attractive alternatives t…

Expanding the Variety of Zirconium‐based Inorganic Building Units for Metal–Organic Frameworks

2019

Two new zirconium-based metal-organic frameworks with the composition [Zr6 O4 (OH)4 (OAc)6 (BDC)3 ] (CAU-26) and [Zr5 O4 (OH)4 (OAc)4 (BDC)2 ] (CAU-27) are reported, which were synthesized from acetic acid, a rarely utilized but green and sustainable solvent (BDC2- : 1,4-benzenedicarboxylate). Structure determination aided by automated electron diffraction tomography revealed that CAU-26 is composed of layers of well-known {Zr6 O8 } clusters interconnected by terephthalate ions. In contrast CAU-27 exhibits a three-dimensional structure with a so far unknown type of one-dimensional inorganic building unit (IBU), which can be rationalized as condensed polyhedron-sharing chains of {Zr6 O8 } cl…
