Synthesis and evaluation of zirconium-89 labelled and long-lived GLP-1 receptor agonists for PET imaging

2020

Contains fulltext : 220838.pdf (Publisher’s version ) (Open Access) INTRODUCTION: Lately, zirconium-89 has shown great promise as a radionuclide for PET applications of long circulating biomolecules. Here, the design and synthesis of protracted and long-lived GLP-1 receptor agonists conjugated to desferrioxamine and labelled with zirconium-89 is presented with the purpose of studying their in vivo distribution by PET imaging. The labelled conjugates were evaluated and compared to a non-labelled GLP-1 receptor agonist in both in vitro and in vivo assays to certify that the modification did not significantly alter the peptides' structure or function. Finally, the zirconium-89 labelled peptide…

Cellular and humoral immune responses against cancer: implications for cancer vaccines.

1991

The key issue in tumor immunology is to identify antigens as target structures for a cancer-selective immunological attack in the tumor-bearing host, resulting in tumor rejection. There is a growing detailed understanding of structural and regulatory gene alterations giving rise to candidate rejection antigens and peptides in tumor cells. As well as reviewing the development of new adjuvant and recombinant vector systems, new approaches are suggested for the construction of cancer vaccines.

Microarray analysis of antibodies induced with synthetic antitumor vaccines : specificity against diverse mucin core structures

2017

Glycoprotein research is pivotal for vaccine development and biomarker discovery. Many successful methodologies for reliably increasing the antigenicity toward tumor-associated glycopeptide structures have been reported. Deeper insights into the quality and specificity of the raised polyclonal, humoral reactions are often not addressed, despite the fact that an immunological memory, which produces antibodies with cross-reactivity to epitopes exposed on healthy cells, may cause autoimmune diseases. In the current work, three MUC1 antitumor vaccine candidates conjugated with different immune stimulants are evaluated immunologically. For assessment of the influence of the immune stimulant on a…

Inverse problem for tripotential measures in the study of buried cavities

1996

This paper presents a solution to the inverse electrical problem for the interpretation of apparent resistivity anomalies due to empty buried cavities of quasi-spherical shape when tripotential measures are carried out. The anomalies of the apparent resistivities ra,rb andrg, and the composed resistivitiesrmand rt were previously calculated for a sufficient class of spherical models of resistivity anomalies. Then, for the whole class of models, some functionals of spatial distribution of the apparent and composed resistivity were identified and analyzed. They represent the average characteristics of the anomalies and, depending in a simple way on the fundamental parameters of the sources of …

Synthesis of aryl azides: A probe reaction to study the synergetic action of ultrasounds and ionic liquids

2011

Abstract The combined effect of ultrasounds and ionic liquids was used to perform the synthesis of aryl azides by nucleophilic aromatic substitution in ionic liquid/[1-butyl-3-methylimidazolium][N 3 ] binary mixtures. The ultrasounds efficiency was analyzed as a function of the substrate and of the ionic liquid structure. In the first case, both 6π and 10π electrons aryl halides were considered. As far as the ionic liquid structure is concerned, both aromatic and aliphatic ionic liquids were taken into account. Among aromatic cations, the effects due to different ability in giving hydrogen bond or π–π interactions were considered. The use of a geminal ionic liquid having an aromatic spacer …

Synthesis of a New Disulfide Affinity Adsorbent for Purification of Human Uterine Progesterone Receptor

2005

For purification of the human uterine progesterone receptor, an affinity adsorbent was synthesized in which the specific ligand (16 alpha-ethyl-3-oxo-19nor-androst-4-ene 17 beta-carboxylic acid) was bound to derivatized celulose using a disulfide-group-containing spacer. The purified receptor protein, isolated by reductive cleavage of the disulfide bond, bound the synthetic gestagen R5020 with high affinity (Kd 12.2 nmol/l). The affinity gel was highly efficient. A 24000-fold purification of progesterone receptor with a recovery of 40% could be achieved in a single step within 6 h. By means of dodecyl sulphate/polyacrylamide gel electrophoresis two main polypeptides with molecular weights o…

A Multiphysics Approach to Constrain the Dynamics of the Altiplano‐Puna Magmatic System

2021

Continuous Interferometric Synthetic Aperture Radar (InSAR) monitoring (> 25 years) has revealed a concentric surface deformation pattern above the Altiplano-Puna magma body (APMB) in the central Andes. Here, we use a joint interpretation of seismic imaging, gravity anomalies and InSAR data to constrain location, 3D geometry and density of the magma body. By combining gravity modelling, thermomechanical modelling, scaling law analysis and Bayesian inference, we are able to create a relationship between the geometry of a mid-crustal magma body and surface observations. Furthermore, we can estimate the uncertainties associated with the geometry of the APMB and identify the most important para…

mTOR Inhibition Improves Antitumor Effects of Vaccination with Antigen-Encoding RNA

2013

Abstract Vaccination with in vitro transcribed RNA encoding tumor antigens is an emerging approach in cancer immunotherapy. Attempting to further improve RNA vaccine efficacy, we have explored combining RNA with immunomodulators such as rapamycin. Rapamycin, the inhibitor of mTOR, was used originally for immunosuppression. Recent reports in mouse systems, however, suggest that mTOR inhibition may enhance the formation and differentiation of the memory CD8+ T-cell pool. Because memory T-cell formation is critical to the outcome of vaccination aproaches, we studied the impact of rapamycin on the in vivo primed RNA vaccine-induced immune response using the chicken ovalbumin-expressing B16 mela…

Encapsulation of in vitro-derived propagules of two genotypes of Capparis spinosa (L.) from Pantelleria Island

2020

Encapsulation technology, combining the advantages of zygotic or gamic seed with those of micropropagation, has recently attracted the interest of researchers as a new propagation approach, mainly due to the unsatisfying results of the traditional propagation strategies. The encapsulation of uninodal microcuttings (3-4 mm long) from two Sicilian Capparis spinosa (L.) genotypes (from Pantelleria Island: 'Tracino' and 'Scauri') was evaluated, observing the influence of the calcium alginate coating and of three different growth regulators (PGRs): 6-benzylaminopurine (BAP), meta-topolin (MT) and zeatin (ZEA), on viability, regrowth and conversion of the propagules. Caper microcuttings were diss…
