Production of soluble eukaryotic recombinant proteins in E. coli is favoured in early log-phase cultures induced at low temperature

Abstract Background Producing recombinant plant proteins expressed in Escherichia coli produce in high yields and in a soluble and functional form can be difficult. Under overexpression conditions, proteins frequently accumulate as insoluble aggregates (inclusion bodies) within the producing bacteria. We evaluated how the initial culture density, temperature and duration of the expression stage affect the production of some eukaryotic enzymes in E. coli. Findings A high yield of active soluble proteins was obtained by combining early-log phase cultures and low temperatures for protein induction. Poly(N-ispropylacrylamide) [PNIPAM] is a widely studied polymer for use in biological applications due to its lower critical solution temperature (LCST) being so close to the human body temperature. Unfortunately, attempts to combine carbon nanotubes (CNTs) with PNIPAM have been unsuccessful due to poor interactions between these two materials. In this work, a PNIPAM copolymer with 1 mol-% pyrene side group [p-PNIPAM] was used to produce a thermoresponsive polymer capable of stabilizing both single and multi-walled carbon nanotubes (MWNTs) in water. Despite their immense potential, the ability to control the dispersion and microstructure of carbon nanotubes remains a hurdle for their widespread use. Stimuli-responsive polymers show conformational changes with an applied external stimulus (pH, temperature, light, etc.). Abstract The coordination compound Fe(BM-4-PTP) 2 (NCS) 2 ⋅2MeOH ( 1 ) including the photoisomerizable ligand BM-4-PTP (1,2-bis(2′-methyl-5′-(pyrid-4″-yl)thien-3′-yl)perfluorocyclopentene) was obtained as an orange powder. The powder turns blue upon photocyclization of the 1,2-bisthienyl photochromic ligand induced by UV light irradiation at room temperature. Photocycloreversion is obtained by visible light irradiation of the material in the solid state. The Efficiency of Polymer Fractionation at Lower Critical Solution Temperatures

The efficiency of a single fractionation step (i.e. of the partition of a given polymer sample between the coexisting phases which form when its solution demixes) is governed by the variation of the mass ratio of the individual P-mers with chain length. These dependences have been measured for the system dimethoxymethane/diethylether/ polystyrene which exhibits lower critical solution temperatures by means of the Baker-Williams method and by means of g.p.c. Collapse of Cylindrical Brushes with 2-Isopropyloxazoline Side Chains Close to the Phase Boundary

A high-molar-mass cylindrical brush polymer with a main chain degree of polymerization of Pw = 1047 is synthesized by free-radical polymerization of a poly-2-isopropyloxazoline macromonomer with Pn = 28. The polymerization is conducted above the lower phase transition temperature of the macromonomer, i.e., in the phase-separated regime, which provides a sufficiently concentrated macromonomer phase mandatory to obtain high-molar-mass cylindrical brushes. Temperature triggered self-assembly of polypeptides into multivalent spherical micelles.

We report herein thermally responsive elastin-like polypeptides (ELPs) in a linear AB diblock architecture with an N-terminal peptide ligand that self-assemble into spherical micelles when heated slightly above body temperature. A series of 10 ELP block copolymers (ELP(BC)'s ) with different molecular weights and hydrophilic-to-hydrophobic block ratios were genetically synthesized by recursive directional ligation. Phases and phase transitions in the mixed molecular system (NaCN)1x(KCN)x

The phase diagram of (NaCN)1−x(KCN)x was examined by neutron powder diffraction in the temperature range 5K ≦T≦300 K. Several non-cubic low-temperature phases were identified for concentrationsx<xc1=0.15 andx≧xc2=0.89. Lattice parameters and ferroelastic deformations were determined from the observed powder patterns. The phase transformations were characterized following the temperature dependence of the appropriate order parameters.
