Sequencing, De Novo Assembly and Annotation of the Colorado Potato Beetle, Leptinotarsa decemlineata, Transcriptome

2012

Background. The Colorado potato beetle (Leptinotarsa decemlineata) is a major pest and a serious threat to potato cultivation throughout the northern hemisphere. Despite its high importance for invasion biology, phenology and pest management, little is known about L. decemlineata from a genomic perspective. We subjected European L. decemlineata adult and larval transcriptome samples to 454-FLX massively-parallel DNA sequencing to characterize a basal set of genes from this species. We created a combined assembly of the adult and larval datasets including the publicly available midgut larval Roche 454 reads and provided basic annotation. We were particularly interested in diapause-specific g…

Pre-stressed translucent panels made of innovative 3D glass components DSC-integrated, for safety and active building façades

2018

The widespread use of glazed envelopes, especially in high-rise buildings, is pushing building designers and researchers to the development of novel solutions for the improvement of their performance. At the University of Palermo, SBskin. Smart Building Skin s.r.l. (an innovative start up and academic spin off) is prototyping precast, pre- stressed and dry-assembled panels made of 3D glass subcomponents, able to reduce the energy consumption of buildings, to produce clean energy and to maintain high level of mechanical resistance against horizontal forces due to winds and earthquakes. The integration of glass subcomponents with third-generation PV technologies -Dye- Sensitized Solar Cells (…

Beyond material factors? Identity, culture and the foreign and security policy of the EU

2015

The evolution of European integration in the field foreign, security and defence policy and the coordination practices established by EU member states cannot be fully explained by looking at material factors only. The chapter adds to the picture the importance of identity, and, in particular, the we-feeling typical of belonging to a security community and security culture in the evolution of the practices adopted for the international projection of the EU and its member states. In consideration of the importance that support to the UN has acquired since the 2003 ESS, both as a strategic priority and as a constitutive element of the EU identity, the chapter focuses on the coordination practi…

The New Models of Public Policy on Early Childhood Education and Care in Spain

2016

Conceptualizing Early Childhood Education and Care (ECEC) as a right for all young children has gained legitimacy in Spain in recent years (Ancheta, 2008). In 2005, the Committee on the Rights of the Child (CRC) stated that young children have specific needs for nurturing, care and guidance (CRC, 2005), and determined that early childhood education should be provided as the right of children to develop personalities, talents and mental and physical abilities from birth (United Nations General Assembly [UNGA], 2011).

Performance comparison of mono-polar and bi-polar configurations of alkaline electrolysis stack through 3-D modelling and experimental fabrication

2020

Abstract Generation of hydrogen using electrolysis process with integrated renewable energy sources is highly important especially in environmental aspects. In this paper, we demonstrate that the enhancement of electrolysis performance of alkaline electrolysis stacks by diminishing the distance between electrodes, while changing the properties of the Membrane Electrode Assembly (MEA). Prior to that, the performances of mono-polar and bi-polar configurations of alkaline electrolysis stack are compared through 3-D modelling and experimental fabrication. At first, two different single cell alkaline electrolysers are designed using SolidWorks as a design software and the designed cell has been …

A membrane-destabilizing peptide in capsid protein L2 is required for egress of papillomavirus genomes from endosomes

2005

ABSTRACT Papillomaviruses are internalized via clathrin-dependent endocytosis. However, the mechanism by which viral genomes pass endosomal membranes has not been elucidated. In this report we show that the minor capsid protein L2 is required for egress of viral genomes from endosomes but not for initial uptake and uncoating and that a 23-amino-acid peptide at the C terminus of L2 is necessary for this function. Pseudogenomes encapsidated by L1 and L2 lacking this peptide accumulated in vesicular compartments similar to that observed with L1-only viral particles, and these mutant pseudoviruses were noninfectious. This L2 peptide displayed strong membrane-disrupting activity, induced cytolys…

Design, manufacturing and testing of a fast disconnecting system for the European target assembly concept of IFMIF

2013

The International Fusion Materials Irradiation Facility (IFMIF) will be equipped with a lithium target assembly to produce the required neutron flux for the irradiation of candidate fusion materials up to a damage rate of 100 dpa (cumulated damage in five years). The present European target assembly design is based on the so called replaceable backplate bayonet concept that was developed with the objective to simplify the maintenance operations for its refurbishment/replacement and to reduce the material for disposal as well. To this purpose it was also conceived to be attached to the lithium pipes and to the beam line by means of remotely operated connections based on clamped flanges with …

Engineering design and steady state thermomechanical analysis of the IFMIF European lithium target system

2013

In the framework of the current IFMIF Engineering Validation and Engineering Design Activities (IFMIF/EVEDA) phase, ENEA is responsible for the design of the European concept of the IFMIF lithium target system which foresees the possibility to periodically replace only the most irradiated and thus critical component (i.e., the backplate) while continuing to operate the rest of the target for a longer period (bayonet backplate concept). In this work, the results of the steady state thermomechanical analysis of the IFMIF EU target assembly are briefly reported highlighting the relevant indications obtained with respect to the fulfillment of the design requirements. © 2013 IEEE.

Epigenomic k-mer dictionaries: shedding light on how sequence composition influences in vivo nucleosome positioning

2014

Abstract Motivation: Information-theoretic and compositional analysis of biological sequences, in terms of k-mer dictionaries, has a well established role in genomic and proteomic studies. Much less so in epigenomics, although the role of k-mers in chromatin organization and nucleosome positioning is particularly relevant. Fundamental questions concerning the informational content and compositional structure of nucleosome favouring and disfavoring sequences with respect to their basic building blocks still remain open. Results: We present the first analysis on the role of k-mers in the composition of nucleosome enriched and depleted genomic regions (NER and NDR for short) that is: (i) exhau…

Tandem repeats lead to sequence assembly errors and impose multi-level challenges for genome and protein databases

2019

AbstractThe widespread occurrence of repetitive stretches of DNA in genomes of organisms across the tree of life imposes fundamental challenges for sequencing, genome assembly, and automated annotation of genes and proteins. This multi-level problem can lead to errors in genome and protein databases that are often not recognized or acknowledged. As a consequence, end users working with sequences with repetitive regions are faced with ‘ready-to-use’ deposited data whose trustworthiness is difficult to determine, let alone to quantify. Here, we provide a review of the problems associated with tandem repeat sequences that originate from different stages during the sequencing-assembly-annotatio…
