Effects of sevoflurane on cognitive deficit, motor function, and histopathology after cerebral ischemia in rats.

2009

Background: The volatile anesthetic sevoflurane exhibits neuroprotective properties when assessed for motor function and histopathology after cerebral ischemia in rats. Damage of hippocampal neurons after ischemia relates to a number of cognitive deficits that are not revealed by testing animals for motor function. Therefore, the present study evaluates cognitive and behavioral function as well as hippocampal damage in rats subjected to cerebral ischemia under sevoflurane compared with fentanyl/nitrous oxide (N 2 O)/O 2 anesthesia. Methods: Thirty-four rats were trained for 10 days using a hole-board test to detect changes in cognitive and behavioral function. Rats were randomly assigned to…

Time course of cerebrovascular autoregulation during extreme Trendelenburg position for robotic-assisted prostatic surgery

2013

Trendelenburg positioning in combination with pneumoperitoneum during robotic-assisted prostatic surgery possibly impairs cerebrovascular autoregulation. If cerebrovascular autoregulation is disturbed, arterial hypertension might induce cerebral hyperaemia and brain oedema, while low arterial blood pressure can induce cerebral ischaemia. The time course of cerebrovascular autoregulation was investigated during use of the Trendelenburg position and a pneumoperitoneum for robotic-assisted prostatic surgery using transcranial Doppler ultrasound. Cerebral blood flow velocity was correlated with arterial blood pressure and the autoregulation index (Mx) was calculated. In 23 male patients, Mx was…

Influence of Ketamine anaesthesia on renal and cardiovascular functions in mongrel dogs

1979

Cardiac minute volume, renal blood flow, pulse volume, heart rate and urinary output were determined by means of experiments made on 10 mongrel dogs. Anaesthesia was maintained by nitrous oxide/oxygen mixture administered with Ketamine. Under the conditions described it was observed that -- contrary to what is found in human patients -- cardiac and renal minute volume as well as pulse volume are significantly decreased. No significant changes has been observed in the heart rate. Whilst urinary output was decreased, the excretion of individual electrolytes was found to be different.

Effects of a single-dose hypertonic saline hydroxyethyl starch on cerebral blood flow, long-term outcome, neurogenesis, and neuronal survival after c…

2012

The beneficial effects of hypertonic saline on neuronal survival and on cerebral blood flow have been shown in several animal models of global and focal brain ischemia. Because of the potential benefits of hypertonic solutions, it is hypothesized that hydroxyethyl starch enhances cerebral blood flow and improves long-term outcome after cardiac arrest and cardiopulmonary resuscitation in an animal model.Laboratory animal study.University animal research laboratory.Fifty-nine male Sprague-Dawley rats.Rats were randomized to receive either 7.2% saline/6% hypertonic saline hydroxyethyl starch (4 mL/kg) or vehicle (NaCl 0.9 %) after 9 mins of asphyxic cardiac arrest and cardiopulmonary resuscita…

No improvement in perfusion and oxygenation of experimental tumors upon application of vasodilator drugs

2001

The oxygen deficiency seen in solid tumors is predominantly caused by an insufficient O2 supply as a result of inadequate tumor perfusion. The aim of this study was to analyze whether a number of vasodilator drugs might be suitable to increase tumor perfusion and consequently improve the oxygenation status of experimental tumors. Rats with s.c. DS-sarcomas were treated with either Na+-nitroprusside (7-25 microg x min(-1) x kg(-1) BW) or nifedipine (10 microg x min(-1) x kg(-1) BW). Red blood cell (RBC) flux was assessed continuously using laser-Doppler flowmetry and mean tumor pO2 was measured polarographically using O2-sensitive catheter electrodes. Systemic application of the vasodilator …

A Scanning Technique to Measure Regional Cerebral Blood Flow and Oxyhemoglobin Level

2001

OBJECTIVE: The application of a laser scanning technique to measure regional cerebral blood flow (CBF) and tissue hemoglobin oxygenation (HbO 2 ) using the rat closed cranial window preparation is described. METHODS: Twenty-nine male Wistar rats were used to consecutively measure local CBF by laser Doppler flowmetry and tissue HbO 2 by a microspectrophotometric method at multiple corresponding cortical locations. The scanning technique used a computer-controlled micromanipulator. Data from three experimental models are presented: the whisker stimulation model, the ischemia-reperfusion model, and the sinus-vein thrombosis model. Sequential changes in local CBF and HbO 2 data before, during, …

Structural adaptations in the murine colon microcirculation associated with hapten-induced inflammation

2007

Objectives: Blood flowing across the vascular endothelium creates wall shear stress, dependent on flow velocity and vessel geometry, that tends to disrupt lymphocyte-endothelial cell adhesion. To identify structural adaptations during acute colitis that may facilitate transmigration, we investigated the microcirculation in a murine model of acute colitis. Methods: In trinitrobenzenesulfonic acid (TNBS)- induced acute colitis, the infiltrating cells and colonic microcirculation was investigated by cellular topographic mapping as well as corrosion casting and 3- dimensional (3D) scanning electron microscopy. Colonic blood velocimetry was performed using intravital microscopy. Results: Clinica…

Microcirculatory alterations in a mongolian gerbil sinus-vein thrombosis model

2001

Summary Background and purpose. The pathophysiology of sinus-vein thrombosis (SVT) is still controversial in patients and experimental animals, the microcirculatory alterations in particular. This study was designed to develop a new sinus-vein thrombosis model and to further elucidate pathophysiological events such as the relationship between local and regional cerebral blood flow and haemoglobin oxygen saturation (HbSO2), changes of the microvasculature, leukocyte behaviour and brain tissue damage. Methods. In a first experimental series, animals were divided into two groups which resulted from different procedures of inducing SVT. In the SSS middle occlusion group (SMO group), SVT was ind…

Cerebral blood flow alterations in a rat model of cerebral sinus thrombosis.

1993

Outcome from sinus vein thrombosis is very variable, with symptoms from headache to coma. Experimental findings suggest that an involvement of cortical veins is necessary to affect the cerebral microcirculation. Laser Doppler flowmetry was used to investigate the regional and temporal changes in local cortical blood flow after experimental occlusion and thrombosis of the superior sagittal sinus and tributary cortical veins in rats. Thrombosis was induced by slow injection of kaolin-cephalin suspension after frontal and caudal ligation of the sagittal sinus in rats. Local cerebral blood flow was measured by laser Doppler flowmetry and correlated with parenchymal damage found 24 hours after i…
