Conservation of the endangered Mediterranean tortoise Testudo hermanni hermanni: The contribution of population genetics and historical demography

2016

International audience; Estimating the genetic variation and demographic trends of species in decline is of major concern in conservation genetics. This study contributes to understanding how historical and anthropogenic factors shape the distribution of current genetic diversity in one of the most endangered reptiles in Western Europe, the Hermann's tortoise Testudo hermanni hermanni. We used 17 microsatellite loci, chosen from a pyrosequencing library specifically developed for the subspecies to genotype eight populations distributed over about 30 sample localities covering almost the entire geographic distribution of the sub-species. The population genetic results reflect a very strong g…

Vertebrate defense against parasites: Interactions between avoidance, resistance, and tolerance

2017

Hosts can utilize different types of defense against the effects of parasitism, including avoidance, resistance, and tolerance. Typically, there is tremendous heterogeneity among hosts in these defense mechanisms that may be rooted in the costs associated with defense and lead to trade‐offs with other life‐history traits. Trade‐offs may also exist between the defense mechanisms, but the relationships between avoidance, resistance, and tolerance have rarely been studied. Here, we assessed these three defense traits under common garden conditions in a natural host–parasite system, the trematode eye‐fluke Diplostomum pseudospathaceum and its second intermediate fish host. We looked at host ind…

Pilot biodiversity assessment of the Hkakabo Razi passerine avifauna in northern Myanmar – implications for conservation from molecular genetics

2019

SummaryThe Hkakabo Razi region located in northern Myanmar is an Important Bird Area and part of the Eastern Himalayan Biodiversity Hotspot. Within the framework of the World Heritage Convention to enlist the site under criterion (ix) and (x), we conducted a biodiversity assessment for passerine birds using DNA barcoding and other molecular markers. Of the 441 bird species recorded, we chose 16 target species for a comparative phylogeographic study. Genetic analysis was performed for a larger number of species and helped identifying misidentified species. We found phylogeographic structure in all but one of the 16 study species. In 13 species, populations from northern Myanmar were genetica…

2017

The endosymbiotic bacterium Wolbachia is the most widespread bacteria in insects, yet the ecology of novel acquisitions in natural host populations is poorly understood. Using temporal data separated by 12 years, I tested the hypothesis that immigration of a parasitoid wasp led to transmission of its Wolbachia strain to its dipteran host, resulting in double-strain infection, and I used geographic and community surveys to explore the history of transmission in fly and parasitoid. Double infection in the fly host was present before immigration of the parasitoid. Equal prevalence of double infection in males and females, constant prevalence before and after immigration in two regions, and inc…

Standardised spider (Arachnida, Araneae) inventory of Lammi, Finland

2020

In June 2019, an ecology field course of the University of Helsinki was held at Lammi Biological Station, Southern Finland. Within this course, the students familiarised themselves with field work and identification of spiders and explored the diversity of species in the area. Three sampling plots were chosen, one in grassland and two in boreal forest, to demonstrate the sampling techniques and, by applying a standardised protocol (COBRA), contribute to a global spider biodiversity project. The collected samples contained a total of 3445 spiders, of which 1956 (57%) were adult. Only adult spiders were accounted for in the inventory due to the impossibility of identification of juveniles. A …

A genetic study on subtropical Anadenanthera colubrina (Vell.) Brenan var. cebil (Griseb.) Altschul tree from Northwestern Argentina

2017

Anadenanthera colubrina (Vell.) Brenan var. cebil (Griseb.) Altschul is a tree species in South America important for its cultural, economic, and medicinal uses. In addition, it represents a trace in memory of the forests that have decreased over the years and for this reason it is not only interesting to study but also important to preserve the tree species for future generations. In this paper, we have characterized the genetic diversity of four populations. We collected seeds from four different sites: San Bernardo (B), El Cebilar (C), Metán (M), and El Gallinato (G) in Salta Province, North Argentina. We then compared the intergenic transcribed sequences of ribosomal DNA, a known geneti…

Genetic Distinctiveness Highlights the Conservation Value of a Sicilian Manna Ash Germplasm Collection Assigned to Fraxinus angustifolia (Oleaceae)

2020

The cosmopolitan genus Fraxinus comprises about 40 species occupying several habitats in the Northern Hemisphere. With some species hybridizing and sharing genetic variants, questions remain on the species assignment of germplasm within the genus Fraxinus despite numerous species-specific assessments. A multidisciplinary approach was employed to provide a definitive insight into the genetics of an endangered Fraxinus &ldquo

Factors affecting somatic embryogenesis in eight Italian grapevine cultivars and the genetic stability of embryo-derived regenerants as assessed by m…

2016

Abstract Embryogenic cultures have been used in cryopreservation, genetic transformation, propagation, virus elimination, induced mutagenesis and in many other biotechnological applications, providing excellent opportunities for biotechnology advances in grapevine. Unfortunately the efficiency of somatic embryogenesis (SE) is genotype-dependent in addition to showing interaction with explant type used and the plant growth regulator (PGR) composition. In order to identify the interaction of these parameters in SE, we tested eight wine grapevine cultivars, three explant types (ovary, anther/filament and stigma/style) and four PGR combinations in a statistically designed experiment. The genoty…

Preserving Biodiversity in Marginal Rural Areas:Assessment of Morphological and Genetic Variabilityof a Sicilian Common Bean Germplasm Collection

2020

The historical cultivation of common bean (Phaseolus vulgaris L.) has resulted in the development of local populations/cultivars in restricted Italian rural areas. Many common bean landraces, still cultivated in small mountain areas from Sicily, have become outdated and endangered due to the commercial varieties spreading. These accessions are poorly known but often represent a genetic heritage to be preserved and enhanced. The ex situ conservation of fifty-seven Sicilian common bean landraces was carried out at the &ldquo
