Background: IMAB362, a chimeric monoclonal antibody that mediates specific killing of cancer cells expressing the tight junction protein Claudin18.2 (CLDN18.2) by activation of immune effector mechanisms, has demonstrated single-agent activity and tolerability in patients ( pts) with heavily pretreated gastric cancer. Methods: Pts with advanced/recurrent gastric and GEJ cancer were centrally evaluated for CLDN18.2 expression by immunohistochemistry (CLAUDETECT® 18.2 Histology Kit). Eligible pts had a CLDN18.2 expression of ≥2+ in ≥40% tumor cells, an ECOG PS of 0–1 and were not eligible for trastuzumab. Pts were randomized 1:1 to first-line EOX (epirubicin 50 mg/m2 and oxaliplatin 130 mg/m2…

Population pharmacokinetic meta-analysis of individual data to design the first randomized efficacy trial of vancomycin in neonates and young infants.

2019

Abstract Objectives In the absence of consensus, the present meta-analysis was performed to determine an optimal dosing regimen of vancomycin for neonates. Methods A ‘meta-model’ with 4894 concentrations from 1631 neonates was built using NONMEM, and Monte Carlo simulations were performed to design an optimal intermittent infusion, aiming to reach a target AUC0–24 of 400 mg·h/L at steady-state in at least 80% of neonates. Results A two-compartment model best fitted the data. Current weight, postmenstrual age (PMA) and serum creatinine were the significant covariates for CL. After model validation, simulations showed that a loading dose (25 mg/kg) and a maintenance dose (15 mg/kg q12h if &lt…

European Guidelines (S1) on the use of high‐dose intravenous immunoglobulin in dermatology

2016

Background The treatment of severe dermatological autoimmune diseases and toxic epidermal necrolysis (TEN) with high-dose intravenous immunoglobulin (IVIg) is a well-established procedure in dermatology. As treatment with IVIg is usually considered for rare clinical entities or severe clinical cases, the use of immunoglobulin is not generally based on data from randomized controlled trials that are usually required for the practice of evidence-based medicine. Owing to the rarity of the indications for the use of IVIg, it is also unlikely that such studies will be available in the foreseeable future. Because the high costs of IVIg treatment also limit its first-line use, the first clinical g…

PIK3CA-related overgrowth spectrum (PROS) encompasses a range of debilitating conditions defined by asymmetric overgrowth caused by mosaic activating PIK3CA variants. PIK3CA encodes the p110α catalytic subunit of phosphatidylinositol-3-kinase (PI3K), a critical transducer of growth factor signaling. As mTOR mediates the growth-promoting actions of PI3K, we hypothesized that the mTOR inhibitor sirolimus would slow pathological overgrowth. Thirty-nine participants with PROS and progressive overgrowth were enrolled into open-label studies across three centers, and results were pooled. For the primary outcome, tissue volumes at affected and unaffected sites were measured by dual energy X-ray ab…

Estimation of the committed radiation dose resulting from gamma radionuclides ingested with food

2014

The objective of the study was to estimate the value of the radiation dose absorbed in consequence of consumption of popular food products for individual age groups. Potatoes, corn and sugar beet were selected for the study. Edible parts of these plants were collected in experimental fields of the KWS Lochow Polska Sp. z o.o. seeding company in Kondratowice (Poland). On the basis of the obtained study results, it can be stated that in consequence of consumption of the selected food products, people may receive increased doses from both natural and artificial radioactive isotopes. The doses calculated for several age groups do not show any health hazards in consequence of consumption of the …

Planting Date and Different N-Fertilization Rates Differently Modulate Agronomic and Economic Traits of a Sicilian Onion Landrace and of a Commercial…

2022

The onion is one of the most widespread bulb vegetables worldwide. Onion cultivation is common in Sicily, as is the use of local genotypes. Cultivation practices are of particular interest in optimizing yield, quality, and profits. The aim of this study was to assess the agronomic response of a Sicilian landrace (Bianca Calda di Comiso, coded L1) and a commercial variety (Bianca di Maggio, coded V1) to different planting dates and N fertilization rates. An economic appraisal was also performed. The two genotypes were assessed using an experimental split-split-plot design with four levels of nitrogen rates (0, 80, 160, and 220 kg N ha−1) and two different planting dates (8 October 2005…

Symptom variability and control in COPD: Advantages of dual bronchodilation therapy

2017

Abstract Background Chronic obstructive pulmonary disease (COPD) is a heterogeneous disorder characterized by usually progressive development of airflow obstruction that is not fully reversible. While most patients will experience symptoms throughout the day or in the morning upon awakening, many patients do not experience their symptoms as constant but report variability in symptoms during the course of the day or over time. Symptom variability adversely affects patients' health status and increases the risk of COPD exacerbations. Methods We examined data from the literature on symptom variability and control in patients with COPD, with focus on the use of inhaled bronchodilator therapy wi…
